摘要:
Nanosized TiO2 with suitable dimensions has been observed to enhance the functions of bone cells; however, the most effective type of nano-TiO2 remains unknown. The Layer-by-Layer (LBL) self-assembly technique has been wildly used to fabricate multilayer thin films of controlled composition, thickness, and architecture. Therefore, to find an appropriate material for orthopedic surgical usage, this study investigates the cytocompatibility and cell activity (using MC-3T3-E1 cells) of nano-TiO2 thin films fabricated by LBL. Four types of nano-TiO2 thin films were synthesized. Surface roughness w measured by field emission scanning electron microscopy (SEM). Hydrophilicity was measured using the contact angle meter. Cell attachment to the materials was observed using SEM. Fluorescent dyes were used to observe the generation of reactive oxygen species and DNA damage. Apoptosis was measured through lactate dehydrogenase leakage assay. The 2-(4,5-dimethyl-2-thiazol-2-yl)-3,5-diphenyl-2H-tetrazolium (MTT) assay was used to determine the viability of the cells attached to the materials. This new film was found to produce better cell proliferation and function; it is comparatively cytocompatible, and therefore may be a better material for orthopedic surgery.

摘要:
As a valuable Chinese herbal medicine, ginseng flower buds are under special physiological phase and have some special medicinal ingredients, which induce that the varieties and properties of its secondary metabolites were extremely complex. Herein, an ideal 2-DE pattern of ginseng flower buds can not be obtained by conventional TCA-acetone extraction method, which has restricted its proteomic studies. Moreover, it is a powerful tool to discuss the synthesis mechanism of ginseng flower buds' secondary metabolites by proteomic technology. In this paper, a three-step extracting procedure of TCA-chloroform/n-butanol-phenol was designed pertinently by comparing TCA-chloroform/n-butanol (4:1) and TCA-phenol method based on the TCA-acetone extraction. Finally, a higher-quality 2-DE pattern was obtained for the foundation of proteomic studies of ginseng buds.

Effects of four antibiotics on induction of lamina calluses and bud differentiation in leaf segments of Populus deltoides I-63x1-69 were studied.The results showed that the rates of bud regeneration were 73.3%, 15.6% and 6.7%, respectively, at 200 mg.L-1 cefotaxim,carbenicillin and streptomycin. However, kanamycin can inhibit the callus induction, bud regeneration and the growth of buds. Therefore, the cefotaxim was optional for the removal of bacterium in popular transformation. Survival rate of buds declined while the concentration of kanamycin increased. Buds could not survive in the medium containing 60 mg.L-1 kanamycin. We suggested that 40 similar to 50 mg.L-1 kanamycin would be optional for genetic engineering of poplar.

摘要:
To investigate the effect of vision clues on spatial cognitive ability of mice, 20 healthy Mus musculus (Km) mice were divided into two groups, control group and test group. During the experiment, the control group was trained in the Morris water maze with the visual signals while all the visual reference objects of Morris water maze were removed when the test group was trained. The latencies and swimming paths of the two groups were recorded for comparing the effect of vision clues on spatial cognitive ability of the mice. The results showed that there was no significant difference of swimming speed between the two groups, but both the latencies to find the hidden platform and swimming paths of the test group increased notably compared with the control group during the place navigation testing and probe trial testing (P<0.05). According to these results, we concluded that vision clues played a critical role on the spatial cognition ability of mice. If the visual reference objects of Morris water maze were removed, the spatial cognitive ability of the mice would decrease, the latencies increased significantly and swimming paths became long and disorder.

The quickly development of bioinformatics based on websites services promoted the study of microbial informatics. Microbial structural genomics and functional genomics try to illuminate the inner relation between the gene structure and function, and to construct gene regulatory networks. The application of Genechip based on bioinformatics became a stronger tool in environmental microbial ecological study and functional enzyme gene orientation. Bioinformatics of web-based services provide a new information platform and technology method for environmental microorganism study.

摘要:
Pichia secretive expression system is one of powerful eukaryotic expression systems in genetic engineering, which is especially suitable for industrial utilization. Because of the low concentration of the target protein in initial experiment, the methods and conditions for expression of the target protein should be optimized according to the protein yield repetitively. It is necessary to set up a rapid, simple and convenient analysis method for protein expression levels instead of the generally used method such as ultrafiltration, purification, dialysis, lyophilization and so on. In this paper, acetone precipitation method was chosen to concentrate the recombinant protein firstly after comparing with four different protein precipitation methods systematically, and then the protein was analyzed by SDS-Polyacrylamide Gel Electrophoresis. The recombinant protein was determined with the feature of protein band by the Automated Image Capture and 1-D Analysis Software directly. With this method, the optimized expression conditions of basic fibroblast growth factor secreted from pichia were obtained, which is as the same as using traditional methods. Hence, a convenient tool to determine the optimized conditions for the expression of recombinant proteins in Pichia was established.

In order to study the cytotoxicity effects of Nano-Fe3O4 Particles on HeLa Cells, 50 mu g/mL, 100 mu g/mL and 200 mu g/mL concentrations of Nano-Fe3O4 were exposed to HeLa cells, twelve hours later, single cell gel electrophoresis (SCGE) was used to detect HeLa cell DNA molecule damage, the content of the malondialdehyde (MDA) were determined by thiobarbiturie acid reactive substance method and the activity of super oxide dismutase (SOD) was determined by xanthine oxldase method. The results shown that under the 50 mu g/mL Nano-Fe3O4 exposure, compared with the control group, neither the tail moment nor the tail DNA% increased (P> 0.05), the increase content of MDA also not significant (P> 0.05), but the reduce activity of SOD is significant (P< 0.05); under the 100 mu g/mL and 200 mu g/mL Nano-Fe3O4 exposure, compared with the control group, the increase of tail moment and tail DNA% both significant (P< 0.05 or P< 0.01), the increase content of MDA also significant(P< 0.05 or P< 0.01), the reduce activity of SOD is significant (P< 0.01). The results suggest that Nano-Fe3O4 could induce the cytotoxicity on HeLa cells.

In order to study the effect of butyl benzyl phthalatee (BBP) on hepatic cells of rat, hepatic cells of rat were exposed to different dose (5 μmol / L, 20 μmol / L, 80 μmol / L) of BBP in vitro for 1 hour. Thiobarbituric acid (TBA) method , KCl-SDS precipitation method and Dinitrophenylhydrazone (DNPH) colorimetric method were used to detect malondialdehyde (MDA) content, DPC coefficient changes and content, respectively. MDA levels in hepatic cells showed a dose-dependent enhancement after BBP exposure. Significant MDA content was observed in all groups compared to control group. The content of carbonyl was all increased in exposure groups, especially in the group of 5μmol / L. DPC coefficient presented a slight increase compared with the control group but no significant difference in the dose of 5 μmol / L and 20 μmol / L. When the concentration of BBP increased to 80 μmol / L, DPC coefficient increased, and there was a significant difference compared with the control group. The carbonyl content was significantly increased in the dose of 5μmol / L. These results suggest that BBP can cause liver damage in rats, and might have toxic effects on the human body.

We investigated the vegetation of sphagnum mire at Erxianyan in Western Hubei Province and analyzed the species composition, association types, dominance and importance value of species. The results show that the species composition and association type of the mire is relatively rich. 111 species of plants, belonging to 85 genera of 51 families, were recorded. The mire vegetation can be divided into 11 associations. Among these, there are 7 Sphagnum-dominated associations. Sphagnum palustre and Juncus setchuensis appears in all of the 7 Sphagnum-dominated associations. The average dominance of Sphagnum in the plots is 90.70%. The importance value of Sphagnum is greater than that of other species in 77.78% sample plots. The results suggested that by human's interference the sphagnum mire at Erxianyan may be regressive succession in two paths: one is to be a community with single dominant species and exiguous species after drainage; the other is to be a weed community with abundant species but no dominant species by intensive interference.

摘要:
Multispectral analysis combined with the Periodic Acid-Schiff method was used to investigate cytological features of insoluble polysaccharides and changes in total insoluble polysaccharide content (TPC) during pollen development in rice, including four cytoplasmic male sterility lines (MSLs) and their corresponding fertility-maintaining lines (FMLs). The multispectral curves of the relative transmittance value (RTV) and the images of developing pollen cells were obtained across a range of successive wavelengths (400–720 nm). A minimum RTV was found near 550 nm indicating an absorption peak of the TPC. Thus, the TPC was measured using the RTV of 550 nm. In the four FMLs, the minimum TPC of developing pollen cells occurred at the late microspore stage, while the maximum TPC occurred at the mature pollen grain stage. The TPC levels of pollen cells were significantly higher in the four FMLs than in their corresponding MSLs during and after pollen abortion. Notably, a steep decrease of multispectral curves at 420 nm appeared before the occurrence of abortion, implying a marker associated with pollen abortion in rice. Our results will be helpful for exploring the changes in TPC during pollen ontogenesis in rice and provide a novel method for the study of bio-macromolecules.

To explore oxidative damage induced by PBDE 209 and its mechanism we carried out this research. Mice were given intragastrically with PBDE 209 via a metal gastric-tube at different levels (0, 50, 100, 200 mg/(kg center dot d)) for seven days and the malondialdehyde (MDA) content, superoxide dismutase (SOD) activities in liver and liver index were mensurated. Results showed that along with elevation of PBDE 209 concentration, SOD activities in mice decreased and 200 mg/(kg center dot d) PBDE 209 exposure group significantly decreased compared with control group (p < 0.01). MDA content first aroused and then decreased. Further more, MDA content in all experimental groups was elevated compared with control group, and 50, 100 mg/(kg center dot d) PBDE 209 exposure group significantly increased (p < 0.01). Liver index of all experimental groups was significantly higher than that of control group (p < 0.01). These results indicate that PBED 209 can induce oxidative damage in liver of mice with apparent dose-dependent relationship.

A bacterial strain named HS-043 with aerobic denitrification character was isolated from wastewater from a textile factory in Wuhan suburb. The denitrification ability of this strain cultured in SC medium and artificial wastewater (AWW) was studied, the results show that the strain HS-043 can removal more than 98% nitrate in artificial wastewater containing appropriate concentration of Fe(2+) and MoO(4)(2-) in 12 hours. According to morphological observation, physiological biochemical identification and sequence analysis of the 16S rDNA, bacterial strain HS-043 was identified as Delftia acidovorans. The aerobic denitrification capacity of D. acidovorans is rarely reported, which means some industrial applications of this novel bacterium.

Organophosphorus (OP) degrading enzymes were widely used in economical and safe detoxification of organophosphorus pesticides from soil, water, food, and aquatic product. In this study, a novel methylparathion degrading enzyme (MPD) was over-expressed in Escherichia coli BL21 (DE3) as a His-tagged fusion protein by the use of lactose as inducer instead of IPTG. SDS-PAGE combined with enzyme activity analysis indicated that lactose-induced expression yield of active MPD was increased nearly 2-fold compared with IPTG as inducer. The optimum temperature and concentration for lactose induction was 37 degrees C and 0.5 % (w/v), respectively. The expressed fusion proteins induced by the two inducers were both purified by Ni-metal-affinity chromatography. The specific activity of the purified recombinant MPD induced by lactose, reaching at 140 mu mol.min(-1).mg(-1), was nearly 0.5-fold higher than that of the purified enzyme induced by IPTG. Thus, lactose was a well alternative inducer to produce active MPD and gave an advantage over enzyme purification. This study suggested a more effective method to produce purified MPD, an ideal enzyme to detoxify OP pesticides for ecosystem restoration.

To study the effect of Butylbenzyl phthalate inducing DNA damages in Rat lung cells. KCl-SDS assay and comet assay were used to reveal the amount of DNA-protein crosslinks and effect of DNA damages induced by different doses of Butylbenzyl phathalate in vitro. The results showed that Butylbenzyl phathalate could cause DNA breakage at low level (10 mu mol/L, P < 0.01), while at higher level (40 mu mol/ L, P < 0.01; 160 mu mol/ L, P < 0.01) it caused crosslinks obviously. In conclusion, higher levels BBP induced significant oxidative damage, and then caused damages to DNA.

CYP51 of Fungi, known as sterol 14-α-demethylase (P450 14DM) is a key enzyme in the biosynthesis of membrane ergosterol. It will lead to the destruction and loss of function of membrane structure and ultimately to fungal death without CYP51. Based on the 5'-end nucleotide sequence of cyp51, the upstream region of cyp51 was obtained by chromosome walking technique to investigate the regulatory mechanism of cyp51 gene expression. The upstream fragment of 490bp was successfully cloned and sequenced. Analyzed by bioinformatics software, the transcription initiation site of cyp51 was predicted by the NNPP software is located at 134bp in front of ATG, while several cis-acting elements are also analyzed by TFSEARCH 1.3 software. The current study was benefit to further investigating the regulation of cyp51 over-expression and molecular mechanism involved in fungicides resistance of Ustilago maydis in future.

The interaction between tri or tetravalent cerium ions and basic fibroblast growth factor (FGF-2) at 0.1-6: 1 molar ratio under physiological condition was studied by fluorescence and CD spectrum. The different spectra alterations of FGF-2 induced by Ce3+ and Ce4+ showed that Ce3+ and Ce4+ caused different conformational changes of FGF-2 respectively, though both of them destabilized the protein. The instability of FGF-2 in the presence of Ce3+ is involved in the oxidation of its free cystein of protein, but that this treatment nearly does not affect the biological activity. As to Ce4+, it not only induced the conformational changes of protein but also inhibits its activity in a dose-dependent manner, which could be relative to the electrostatic repulsion between Ce4+ and its basic amino acid residues (pI=9.6) or the specific binding of Ce4+ to deprotonated amino acid residues. The interesting results would be helpful to investigate the problem of the stability of proteins.

To investigate the oxidative damage induced by di-(2ethylhexyl) phthalate (DEHP) and analyze the possible function of such effects. This study took Esisenia foelide as experimental material to detect the levels of DNA-protein crosslinks (DPC), the contents of malondialdehyde (MDA), and the activity of surperoxide dismutase (SOD). The Esisenia foelide were exposed to DEHP for 2 hours in vivo with the bath exposure technique. DPC coefficient increases along with the concentration of DEHP (0.2 mmol center dot L-1, 0.4 mmol center dot L-1, 0.8 mmol center dot L-1, 1.6 mmol center dot L-1) rising, and it can cause DPC significantly (p < 0.01) compared with the control group, but the contents of MDA did not change significantly with the increase of DEHP concentrations compared with the control group (p > 0.05). A decrease in the activities of SOD was observed. Significant changes of SOD activity could be observed in 0.4, 0.8 and 1.6 mmol center dot L- 1 (p < 0.01) groups. To conclusion, high levels DEHP induced significant oxidative damage, then caused damage to DNA.

Cadmium, as an inorganic pollutant, its toxic effects on mammals were widely studied, while less attention was paid on its endocrine disrupting effects. In this paper, the effects of CdCl2 on mice uterine-ovary wet weight, modulation of ER (estrogen receptors) mRNA level and their relation were studied. It showed that uterus-ovary wet weight, the expression of ER alpha (alpha subtype) and ER beta (beta subtype) were of significant difference in 930 mu g/kg.bw CdCl2 dosing group compared with the control. However, only up-regulation of ER beta. was detected in 9.3 mu g/kg.bw CdCl2 dosing group. It suggested that the high expression of ER beta might protect mice from increasing of uterus-ovary wet weight. For CdCl2 estrogenic activity, the ER expression mechanism and ER-responsive elements pathway need further examination.
