摘要:
An effective multi-residue pretreatment technique, solid-phase extraction (SPE) combined with dispersive liquid-liquid microextraction (DLLME), was proposed for the trace analysis of 14 polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) in milk samples using gas chromatography-mass spectrometry (GC-MS). Interesting analytes in milk samples were extracted with hexane after protein precipitation. The hexane extracts were loaded on an LC-Florisil column to isolate analytes from the milk matrix. The elutes were dried and dissolved in acetone, which was used as the disperser solvent in subsequent DLLME procedures. The effects of several important parameters on the extraction efficiency were evaluated. Under the optimized conditions, a linear relationship was obtained in the range of 0.02-10.00 μg/L (PCBs) and 0.5-100.00 μg/L (PBDEs). The LOD (S/N=3) and relative standard deviations (RSDs, n=5) for all analytes were 0.01-0.4 μg/L and 0.6-8.5%, respectively. The recoveries of the standards added to raw bovine milk samples were 74.0-131.8%, and the repeatabilities of the analysis results were 1.12-17.41%. 摘要:
A highly enantioselective organocatalytic intermolecular conjugate addition of oximes to β-nitroacrylates has been developed. The highly functionalized adducts obtained are valuable precursors for asymmetric synthesis, as demonstrated by the synthesis of β2,2-amino acids and oxazolidin-2-ones. © 2010 American Chemical Society.

摘要:
The scope of the novel ruthenium-catalyzed tandem cross-metathesis/ intramolecular-hydroarylation sequence is described. This methodology offers a practical and efficient synthesis of structurally diverse and complex tetrahydrocarbazoles in good to excellent yields (up to 98%). Moreover, preliminary efforts towards the development of an enantioselective version of the current process by sequential catalysis with ruthenium complex and chiral amine are presented, with high yields and enantioselectivities (up to 88% yield and 91% ee). © 2010 Wiley-VCH Verlag GmbH&Co. 摘要:
Palladium-catalyzed cyclocarbonylations of 2-iodoanilines with various terminal alkynes have been carried out by the use of commercially available molybdenum hexacarbonyl as a convenient and solid carbon monoxide source. The reactions were conducted at 160 °C for 30 min under microwave irradiation and in the presence of Et3N in THF, affording the corresponding 2-quinolone derivatives in good regioselectivities and yields.

摘要:
An efficient, one-pot, multicomponent reaction of aldehydes 1, p-methoxyaniline (2a), and 2-vinylindoles 3 was developed. This approach provides a practical approach to synthetically and biologically significant tetrahydro-γ-carboline and tetrahydroquinoline derivatives in good yields through a chemo- and regioselectivity switch, which can be tuned by simply changing the substituent on the indole component under identical reaction conditions. © 2010 Wiley-VCH Verlag GmbH & Co. (Chemical equation presented) A change in tune: A novel [4+1] annulation between atropisomeric sulfur ylides 1 and unsaturated imines 2 was developed. The method allows the synthesis of enantioenriched pyrroline products 3 in high yield and with excellent stereocontrol. 摘要:
A new cyclometalating bridging ligand 1,3,6,8-tetra(2-pyridyl)pyrene was designed and synthesized through 4-fold Suzuki couplings between 1,3,6,8-tetrabromopyrene and 2-pyridylboronate. A bis-cyclometalated bisruthenium complex bridged by this ligand showed the presence of an electronic coupling between individual metal centers, as indicated by electrochemical and spectroscopic studies.

摘要:
Asymmetric Michael addition reactions of aldehydes to nitroolefins have been successfully initiated by a series of primary amine thiourea bifunctional catalysts, with high enantioselectivities (90–98% ee) and excellent yields (80–96%). The privileged quinine scaffold was found to be essential to the reaction efficiency and enantioselectivity.

Ustilago maydis infection is a serious disease affecting corn crops worldwide. Sterol 14α-demethylase (CYP51) is one of the key enzymes of sterol biosynthesis and an effective target of antifungal drugs. To further study the interaction between CYP51 and drugs and exploit more specific 14α-demethylase inhibitor (DMI) fungicides for U. maydis, in this study homology modeling of CYP51 from U. maydis (UmCYP51) templated as the eukaryotic orthologues (the human CYP51) and screening of new DMI fungicides based on optimized expression were carried out for the first time. In addition, XF-113 and ZST-4 were screened by analyzing the spectral characteristics between the purified UmCYP51-35 and fungicides. These results provide a theoretical basis and new ideas for efficient design and development of new antifungal drugs. ©2010 American Chemical Society.

BACKGROUND: Sterol 14α-demethylase (CYP51), a key target of azole (DMI) fungicides, can be expressed in both prokaryotes and eukaryotes. Green mould of citrus, caused by Penicillium digitatum (Pers.) Sacc., is a serious post-harvest disease. To develop specific and more effective fungicides against this disease, the characteristics of the interaction between sterol 14α-demethylase from P. digitatum (PdCYP51) and possible new fungicides were analysed. The cyp51 gene of P. digitatum was cloned and expressed under different conditions in Escherichia coli (Mig.) Cast. & Chalm., and the binding spectra of PdCYP51 were explored by the addition of two commercial azoles and four new nitrogen compounds.RESULTS: The yield of soluble protein (PdCYP51) was largest when expressed in Rosetta (DE3) induced by 0.5 mM IPTG for 8 h at 30 °C. Compound B (7-methoxy-2H-benzo[b][1,4]thiazine-3-amine) showed the strongest binding activity of the four new nitrogen compounds, with a K d value of 0.268 μM. The K d values of the six compounds were significantly correlated with their EC 50 values.CONCLUSION: The spectral analysis and bioassay results could be used to screen the new chemical entities effectively. Compound B, selected by virtual screening from a commercial chemical library, is a candidate for a new DMI fungicide. These results provide a theoretical basis and new ideas for efficient design and development of new antifungal agents. 摘要:
A nitrogen-containing compound, named methyl-4-phenyl-2-(2,4,6-triisopropyl-phenylsulfonamido)but-3-enoate (MPSN), is used as a precursor for inverse-electron-demand aza-Diels-Alder reaction. Its 1H and 13C nuclear magnetic resonances (NMR) were fully assigned by a combination of one- and two-dimensional NMR experiments. It was found that this compound had two conformers (a and b) in the process of assignments. Exchange spectroscopy (EXSY) was used to study the exchange dynamics of this compound in solvent. On the basis of 1H NMR and EXSY experiments at various temperatures, thermodynamic and dynamic parameters of the conformational exchange process were deduced for this compound. The free-energy activation (ΔG≠) at 298 K is 15.07 ± 0.6 kcal mol-1, which suggests that the conformational exchange might be the result of C=N bond rotation. © 2010 Springer.

A series of thiourea-amine bifunctional catalysts have been developed by a rational combination of prolines with cinchona alkaloids, which are connected by a thiourea motif. The catalyst 3a, prepared from L-proline and cinchonidine, was found to be a highly efficient catalyst for the conjugate addition of ketones/aldehydes to a wide range of nitroalkenes (up to 98/2 dr and 96% ee). The privileged cinchonidine backbone and the thiourea motif are essential to the reaction activity and enantioselectivity.

Simply complex: Diverse and structurally complex chroman derivatives with a quaternary stereocenter have been obtained through the titled reaction of thiols with nitroolefin enoates using the bifunctional catalyst 1. [Chemical equation presented] Under control: A format [4+1]/[3+2] cycloaddition cascade of sulfur ylides and alkene-tethered nitroolefins has been developed, and provides an efficient synthesis of fused polycyclic heterocyclic compounds in good to excellent overall yields with excellent diastereocontrol (see scheme). A catalytic asymmetric variant of this reaction has also been disclosed © 2009 Wiley-VCH Verlag GmbH & Co. KCaA.

摘要:
An efficient and novel organocatalyst has been developed for the asymmetric desymmetrization of meso- and prochiral ketones by direct Michael addition to nitroolefins. This strategy can afford the desymmetrization products with excellent diastereo- (up to >99:1) and enantioselectivity (up to 96%) in great yields (up to 95%). (C) 2009 Elsevier Ltd. 1H-1,2,4-Triazole reacted with 2-butenal in the presence of diaryl prolinol silyl ether 3 and benzonic acid to give 3-(1H-1,2,4-triazol-1-yl)butanal 4, which was subsequently reduced and then treated with various acyl chloride to generate enantioriched 3-(1H-1,2,4-triazol-1-yl)butyl benzoates 6. Some of triazoles 6 exhibited strong binding interactions with the cytochrome P450-dependent sterol 14α-demethylase (CYP51). For example, compound (R)-6f showed the best binding activity with Kd 0.3381 μM. © 2009 Elsevier Ltd. All rights reserved.

摘要:
t-BuOK was found to be an effective promoting reagent for tandem ring-opening/closing reactions of various N-Ts aziridines and aryl propargyl alcohols to afford dihydroxazine derivatives in moderate to good yields. A plausible reaction mechanism has been proposed.
