摘要:
Alternating benzene-heteroaryl oligomers possess fascinating optoelectronic properties and a wide range of applications. This article presents the facile synthesis of benzenepyrrole oligomers with diverse functional groups and the elongated alternating heterocycle-benzene-pyrrole oligoaryls. The syntheses are based on a one-pot, three-step reaction of propargylic dithioacetals and imines. The subtle influence of functional groups, such as ether, ester, hydroxy, and dithiacetal groups on the peripheral benzene rings, on the reaction, has been explored and the reaction conditions were finely changed accordingly to achieve reasonable yields. The elongation could be accomplished by using a similar procedure. The fluorescent properties of original and elongated alternating benzene-pyrrole oligomers were determined. Theoretical calculations preliminarily reveal that the fluorescent intensity is closely related to the molecular geometry and to the HOMO/LUMO energy difference in the ground state. © 2011 Wiley-VCH Verlag GmbH & Co. 摘要:
利用α-硫辛酸、生物素、长醚链二胺等原料,用柱色谱进行纯化和分离,设计合成了标题化合物,并用1HNMR、GC-MS、IR等方法对中间体及目标产物的分子结构进行了表征。通过改变生物素分子中的侧链部分,可合成不同种类的生物素衍生物,增加了生物素-亲合素/链霉亲合素/中性亲合素自组装膜体系的材料的种类,此自组装膜体系在分子器件、生物与化学传感器、生物膜等很多方面有着广泛的应用。

摘要:
An electrochemical strategy on the basis of rationally designed molecularly imprinted sol-gel polymer embedded with gold nanoparticles (AuNPs) is developed for the specific and sensitive determination of Sudan I. The rationally designed sensing Sudan I imprinted sol-gel was prepared by mixing Sudan I with 3-aminopropyltriethoxysilane, tetraethoxysilane, chitosan, and AuNPs, followed by copolymerization and extraction of the template molecules. The hybrid forming membrane was characterized by SEM and FTIR-ATR, and used for the linear sweep voltammetric (LSV) determination of Sudan I in water/ethanol solutions. The LSV responses exhibited high sensitivity and selectivity, as discriminated from Sudan I analogues. Under optimal experimental conditions, LSV peak currents were linearly proportional to the concentrations of Sudan I in the range from 0.1 ×10<sup>-7</sup> to 1.0 ×10<sup>-5</sup> M, with a detection limit of 2.0 ×10<sup>-9</sup> M. The strategy is generally applicable in developing sensitive, selective, and moreover, reusable electrochemical sensors for quantitative determination of electroactive species. ©2011 Elsevier B.V. 摘要:
通过嘧啶甲酰氯与取代的苄胺肟反应得到一系列结构新颖的5-嘧啶基-1,2,4-噁二唑类化合物,并对其结构进行了表征,培养并测定了化合物5a的晶体结构;将目标化合物晶体结构对接到靶酶酵母乙酰辅酶A羧化酶(ACCase)的活性位点,发现目标化合物与复合物晶体中的禾草灵分子的构象及结合模式相似;初步生物活性测试表明部分化合物具有较好的除草活性.

摘要:
A new sandwich-like electrochemical immunosensor has been developed for quantification of organophosphorylated acetylcholinesterase (OP-AChE), an exposure biomarker of organophosphate pesticides and nerve agents. Zirconia nanoparticles (ZrO2 NPs) were anchored on a screen printed electrode (SPE) to preferably capture OP-AChE adducts by metal chelation with phospho-moieties, which was selectively recognized by lead phosphate-apoferritin labeled anti-AChE antibody (LPA-anti-AChE). The sandwich-like immunoreactions were performed among ZrO2 NPs, OP-AChE and LPA-anti-AChE to form ZrO2/OP-AChE/LPA-anti-AChE complex and the released lead ions were detected on a disposable SPE. The binding affinity was investigated by both square wave voltammetry (SWV) and quartz crystal microbalance (QCM) measurements. The proposed immunosensor yielded a linear response current over a broad OP-AChE concentrations range from 0.05nM to 10nM, with detection limit of 0.02nM, which has enough sensitivity for monitoring of low-dose exposure to OPs. This method avoids the drawback of unavailability of commercial OP-specific antibody as well as amplifies detection signal by using apoferritin encoded metallic phosphate nanoparticle tags. This nanoparticle-based immunosensor offers a new method for rapid, sensitive, selective and inexpensive quantification of phosphorylated adducts for monitoring of OP pesticides and nerve agents exposures. © 2011 Elsevier B.V.

基于新型作用靶标开展新型除草剂的合理设计与合成,对于创制除草剂新品种具有重要意义。近年来文献报道D1蛋白酶可作为除草剂的新型作用靶标,依据D1蛋白酶活性腔结构特征和前期工作获得的咪唑啉-2,4-二酮、喹唑啉-4(3H)-酮、苯并[d][1,3]噁嗪-2-酮的先导结构信息,通过分子对接和物化性质计算,从源头开展对先导结构信息化合物的分子结构优化,合成具有与D1蛋白酶活性腔结构匹配的、三角拓扑结构骨架的咪唑啉-2,4-二酮、喹唑啉-4(3H)-酮、苯并[d][1,3]噁嗪-2-酮三大类化合物。对全部化合物进行活体生物活性测定和基于D1蛋白酶的荧光极化法高通量筛选;采用高效液相法和表面等离子体共振技术进一步研究部分化合物的生物活性机制及其与D1蛋白酶的相互作用动力学。通过本项目工作的实现,预期获得对D1蛋白酶作用机制明确的新型除草剂先导结构化合物。

摘要:
Aryloxyphenoxypropionates (APPs) are a class of herbicides targeting acetyl-CoA carboxylase (ACCase) in monocot chloroplast. The article presents the rational design and synthesis of a series of APPs with 2- and 4-pyrimidinyloxy moieties. The developed synthetic protocols are versatile and allow convenient functionalization of the phenoxypropionate core with various 2- or 4-pyrimidinyl moieties via the oxygen bridge by taking advantage of the reactivity difference between 2-methylsulfonyl and 4-chloro on the pyrimidine ring. Preliminary bioassay shows that most of title compounds have good herbicidal activities against rape and barnyard grass at 100 mg/L. Several compounds are more potent to inhibit the stalk growth of barnyard grass than the commercial herbicide cyhalofop. © ARKAT USA, Inc.

植物细胞内囊体中的D1蛋白酶是近年来发现的潜在的除草剂作用靶标.以异噁唑甲酰赖氨酸脲先导结构信息为基础,设计合成了未见报道的异噁唑甲酰赖氨酸类化合物11个.目标化合物分子结构经IR,1HNMR,元素分析予以确证.平皿法活体生物活性测试表明部分化合物具有中等除草活性;化合物8h的离体生物活性测试表明其对D1蛋白酶有一定的抑制作用.

摘要:
The labeling strategy with gold nanoparticles for the conventional surface plasmon resonance (SPR) signal enhancement has been frequently used for the sensitive determination of small molecules binding to its interaction partners. However, the influence of gold nanoparticles with different size and shape on SPR signal is not known. In this paper, three kinds of gold nanoparticles, namely nanorods, nanospheres, and nanooctahedrons with different size, were prepared and used to investigate their effects on the conventional SPR signal at a fixed excitation wavelength 670 nm. It was found that the SPR signal (i.e., resonant angle shift) was varied with the shapes and sizes of gold nanoparticles in suspension at a fixed concentration due to their different plasmon absorbance bands. For gold nanorods with different longitudinal absorbance bands, three conventional SPR signal regions could be clearly observed when the gold nanorod suspensions were separately introduced onto the SPR sensor chip surface. One region was the longitudinal absorbance bands coinciding with or close to the SPR excitation wavelength that suppressed the SPR angle shift. The second region was the longitudinal absorbance bands at 624 to 639 and 728 to 763 nm that produced a moderate increase on the SPR resonant angle shift. The third region was found for the longitudinal absorbance bands from 700 to 726 nm that resulted in a remarkable increase in the SPR angle shift responses. This phenomenon can be explained on the basis of calculation of the correlation of SPR angle shift response with the gold nanorod longitudinal absorbance bands. For nanospheres and nanooctahedrons, the SPR angle shift responses were found to be particle shape and size dependent in a simple way with a sustaining increase when the sizes of the nanoparticles were increased. Consequently, a guideline for choosing gold nanoparticles as tags is suggested for the SPR determination of small molecules with binding to the immobilized interaction partners.

The identification of aroma compounds, formed from the reactions of l-ascorbic acid with l-threonine/l-serine at five different pH values (5.00, 6.00, 7.00, 8.00, or 9.55) and 143 ± 2 °C for 2 h, was performed using a SPME-GC-MS technique, and further use of LRI. The results showed 35 aroma compounds. The reaction between l-ascorbic acid and l-threonine/l-serine led mainly to the formation of pyrazines. Many of these were alkylpyrazines, such as 2-methylpyrazine, 2,5-dimethylpyrazine, 2-ethylpyrazine, 2-ethyl-6-methylpyrazine, 2-ethyl-5-methylpyrazine, 3-ethyl-2,5-dimethylpyrazine, 2,3-diethyl-5-methylpyrazine, and 3,5-diethyl-2-methylpyrazine; other compounds identified were furans and aldehydes. More volatiles were generated in l-ascorbic acid with l-threonine systems than in l-ascorbic acid with l-serine systems. The studies showed that furans, such as furfural, 2-furanmethanol, benzofuran, 2,5-furandicarboxaldehyde and 2-furfurylfuran were formed mainly at acidic pH. In contrast, higher pH values could promote the production of pyrazines. © 2009 Elsevier Ltd. All rights reserved.

摘要:
A facile, one-step synthesis of nanocomposites using multiwalled carbon nanotube coating gold nanoparticles (MWCNTs-Au) was presented. Scanning electron microscopy and UV-vis spectroscopy confirmed that more than 97% of gold nanoparticles have been loaded on the surface of carbon nanotubes without congregation. The formed MWCNTs-Au nanocomposites offered an extremely hydrophilic surface for biomolecule adhesion, leading to a stable acetylcholinesterase (AChE) biosensor. Due to the excellent conductivity of the nanocomposites, the immobilized AChE showed favorable affinity to acetylthiocholine (ATCl) and could catalyze the hydrolysis of ATCl with a K<inf>m</inf><sup>app</sup> value of 268 μM to form thiocholine, which was then oxidized to produce a detectable and fast response. Based on the inhibition of organophosphates (OPs) on the enzymatic activity of AChE, the magnitude of peak current from thiocholine on the biosensor is a simple and effective way to biomonitoring of OPs exposure. Using malathion as a model compound, the inhibition of malathion was proportional to its concentration ranging from 1.0 to 1000 ng mL<sup>-1</sup> and from 2 to 15 μg mL<sup>-1</sup>, with a detection limit 0.6 ng mL<sup>-1</sup>. The developed biosensor exhibited good reproducibility and acceptable stability, thus providing a new promising tool for analysis of enzyme inhibitors. ©2009 Elsevier B.V. All rights reserved.

摘要:
A simple method to immobilize acetylcholinesterase (AChE) on polypyrrole (PPy) and polyaniline (PANI) copolymer doped with multi-walled carbon nanotubes (MWCNTs) was proposed. The synthesized PAn-PPy-MWCNTs copolymer presented a porous and homogeneous morphology which provided an ideal size to entrap enzyme molecules. Due to the biocompatible microenvironment provided by the copolymer network, the obtained composite was devised for AChE attachment, resulting in a stable AChE biosensor for screening of organophosphates (OPs) exposure. MWCNTs promoted electron-transfer reactions at a lower potential and catalyzed the electro-oxidation of thiocholine, thus increasing detection sensitivity. Based on the inhibition of OPs on the AChE activity, using malathion as a model compound, the inhibition of malathion was proportional to its concentration ranging from 0.01 to 0.5. and mu;g/mL and from 1 to 25. and mu;g/mL, with a detection limit of 1.0. ng/mL. The developed biosensor exhibited good reproducibility and acceptable stability, thus providing a new promising tool for analysis of enzyme inhibitors. © 2010 Elsevier B.V.

摘要:
The reaction of l-ascorbic acid with l-cysteine in heated aqueous solution (141 ± 1 °C) at five different pH values (5.00, 6.00, 7.00, 8.00, or 9.00) for 2 h, resulted in the formation of a complex mixture of aroma volatiles. The volatile compounds generated were analysed by SPME-GC-MS. The results gave 43 aroma compounds. The reaction between l-ascorbic acid and l-cysteine led mainly to the formation of alicyclic sulphur compounds, thiophenes, thienothiophenes, thiophenones, thiazoles and pyrazines, most of which contain sulphur. Many of these volatiles had meaty flavour. The origin of many of the compounds was explained. The studies showed that thienothiophenes and thienones were formed mainly at acidic pH. In contrast, higher pH values could promote the production of thiophenes, thiazoles and pyrazines. © 2010 Elsevier Ltd. All rights reserved.

摘要:
This work described a highly sensitive amperometric biosensor for organophosphates (OPs) pesticides based on immobilization of acetylcholinesterase (AChE) on multiwall carbon nanotubes (MWCNTs)-β-cyclodextrin (β-CD) composite modified glassy carbon electrode. The synthesized composite through polymer wrapping strategy has been characterized by attenuated total reflection Fourier-transform infrared spectra (ATR-FTIR) measurement and scanning electron microscopy (SEM) images. Due to the good dispersibility and porous structures of MWCNTs-β-CD composite, the resulting surface provided a favorable microenvironment for acetylcholinesterase biosensor fabrication and maintained the bioactivity of AChE for screening of OPs exposure. MWCNTs promoted electron-transfer reactions at a lower potential and catalyzed the electro-oxidation of thiocholine, thus increasing detection sensitivity. Based on the inhibition of OPs on the AChE activity, using dimethoate as a model compound, the inhibition of dimethoate was proportional to its concentration ranging from 0.01 to 2.44 and 2.44 to 10.00 μM, with a detection limit was 2 nM (S/N = 3). The developed biosensor exhibited good reproducibility and acceptable stability, thus providing a new promising tool for analysis of enzyme inhibitors. ©2010 Elsevier B.V. All rights reserved.

In the title molecule, C26H24N4O 2S, the dihedral angle between the isoxazole ring and the adjoining benzene ring is 21.4 (5)°, and between the isoxazole ring and the thia-zole ring is 14.3 (4)°. The piperidine ring is in a chair conformation. In the crystal structure, molecules are linked by inter-molecular N - H⋯O and weak C - H⋯O hydrogen bonds into one-dimensional chains along [001].
