含嘧啶环的新型黄酮类化合物的合成及生物活性研究

摘要:
黄酮类化合物是一类植物次级代谢产物,广泛存在于自然界中,他的种类繁多,包含查尔酮,黄烷酮,黄酮,黄酮醇等[1,2].由于其具有广泛的生理作用,如抗氧化性,抗癌,抗炎,杀菌等[3-5],因而近年来,引起了化学工作者的极大兴趣,本文设计将嘧啶杂环这种高活性因子的基团,引入到黄酮类化合物的骨架结构中,以期望得到更高活性的一系列化合物.本文利用生物电子等排法,用嘧啶环替代苯环,设计、合成了一系列含有嘧啶环的新型黄酮类化合物,包含2-羟基查尔酮,黄烷酮,黄酮.如下图所示,将4,6-二羟基嘧啶和POCl3,N,N-二甲基甲酰胺发生vilsmeier反应得到4,6-二氯-5-嘧啶甲醛1;然后在室温条件下将4,6-二氯-5-嘧啶甲醛和邻羟基苯乙酮,以氢氧化钠为碱,在质子性溶剂醇的作用下发生羟醛缩合反应得到2-羟基查尔酮2;最后,把2-羟基查尔酮作为一种活泼中间体,在醋酸钠、乙醇的条件下,回流得到黄烷酮3,或在二氧化锡、二甲亚砜的条件下,回流得到黄酮4.所合成化合物的结构经1H NMR、13C NMR、MS及IR确证,并利用DPPH法[6]进行了体外抗氧化活性初筛.初步的抗氧化活性测试表明,有6个化合物对DPPH自由基的清除率达到90%以上,其活性接近于阳性对照维生素C(Vc).构效关系分析显示,A环上3和4位都有羟基的化合物(2j,3j,4j),都具有很好的清除DPPH自由基的活性,在2 mmol/L时对DPPH的清除均可达95%以上.

摘要:
A novel compound of 1,2-dis(1,3-diphenylpropan-2-yl)disulfane (C 30H30S2, Mr = 454.27) was synthesized in ethanol with 1,3-diphenyl-2-propanone as raw materials, giving high yield of product (90%). Its structure was determined by IR, 1H NMR, MS, elemental analysis and X-ray diffraction. The crystal belongs to monoclinic, space group C2 with a = 18.727(3), b = 6.5179(9), c = 13.7576(18) Å, β = 131.0610(10)°, V = 1266.2(3) Å3, Z = 2, Dc = 2.136 g/cm3, μ = 1.078 mm-1, F(000) = 843, R = 0.0490 and wR = 0.1247 for 3211 observed reflections with I > 2σ(I). X-ray analysis reveals that the molecule is not symmetrical. There exist no classical hydrogen bonds and π-π stacking interactions. Moreover, the thermal stability of compound 3 was investigated. 利用苄硫醇、溴十一酸、4,7,10-三氧-1,13-十三烷二胺等原料,通过活化酯法和NH3/Na还原脱苄基,合成了一种含巯基的生物素衍生物——11-巯基十一酸-(13-生物素酰胺基-4,7,10-三氧十三烷基)酰胺,并用1 H NMR、IR对产物的分子结构进行了表征.11-巯基十一酸-(13-生物素酰胺基-4,7,10-三氧十三烷基)酰胺可以在金表面自组装,而自组装膜在分子器件、生物与化学传感器、生物膜等很多方面有着广泛的应用.

摘要:
Diphenylmethane is an important substructure unit that is found not only in a number of natural products[1], but also in many synthetic compounds of biological and pharmaceutical importance[2]. Significant efforts are devoted to the construction of this motif and its derivatives, and diverse synthetic methods, roughly classified as conventional[3] and catalytic ones[4], have been reported so far. In this article, we described a first example for the synthesis of symmetrical diarylmethanes by facile, one-pot, two-step, palladium-catalyzed sequential coupling(Scheme 1). In the facile one-pot reaction, aryl methyl ketone acts in formal as the methylene donor. The experimental facts, including TLC monitoring, speculated intermediates as the raw materials, ex situ IR spectroscopy analysis of the co-product cesium benzoate, as well as the cross-coupling reactions of two different aryl halides, indicate a mechanism involving a palladium-catalyzed sequential two-step coupling process. The reaction is applicable for a broad spectrum of substrates(up to 21 examples) in good to excellent yields. The access to unsymmetrical diarylmethanes with this method is also explored and various factors are discussed.

摘要:
含氮杂环类化合物是新农药创制研究的热点之一[1].苯并噁嗪酮及苯并嘧啶酮类化合物作为具有多样生物活性的重要杂环化合物,有其广泛的应用,如杀菌剂、除草剂、杀虫剂、抗癌、镇静剂、拮抗剂等,因而被广泛地应用于农药医药等领域[2-5].苯氧乙酸类化合物也是一种很好的生长素抑制剂,易被植物的根、叶吸收.这类除草剂被植物吸收后,体内的生长素的浓度高于正常值,从而打破了植物体内的激素平衡,影响到植物的正常代谢,导致敏感杂草的一系列生理生化变化,组织异常和损伤.本文采用活性亚结构拼接法,将苯并噁嗪酮与苯氧乙酸类结构进行拼接,设计并合成了一系列新型2-苯氧甲基-4H-1,3-苯并噁嗪-4-酮类衍生物,并以此为中间体通过亲核取代反应,合成了2-苯氧甲基-3H-苯并嘧啶-4-酮衍生物.如Scheme 1所示,首先将苯氧乙酸1和SOCl2一起加热回流转变为酰氯2,然后在三乙胺和DMF的条件下,将其与邻氨基苯甲酸进行一锅煮关环反应得到中间体3.随后将3和水合肼溶液、甲胺醇溶液、醋酸铵与甘氨酸固体在醋酸中加热回流得到2-苯氧甲基-3H-苯并嘧啶-4-酮衍生4.所合成化合物的结构经1H NMR、13C NMR、MS及IR确证,并采用行业标准农药室内除草剂活性测试平皿法(NY/T 1155.1-2006)进行了室内除草活性初筛.初步的除草活性测试表明,这些化合物都有很好的除草活性,例如,在浓度为1 ppm时,化合物3a,对油菜的抑制率可以达到92%,化合物4a-I,4b-I,4d-I对油菜的抑制率可以达到87%以上.且它们具有和生长素性抑制剂苯氧乙酸类化合物有类似的的抑制表型.

摘要:
A novel palladium-catalyzed coupling reaction of an aryl methyl ketone with two molecules of an aryl halide to yield symmetric diarylmethanes is described. In the facile one-pot reaction, the aryl methyl ketone acts as a formal methylene donor. The experimental facts, including TLC monitoring, speculated intermediates as the raw materials, analysis of the cesium benzoate coproduct by ex situ IR spectroscopy, and the cross-coupling reactions of two different aryl halides, indicate a mechanism involving a palladium-catalyzed sequential two-step coupling process, in which the presence of a trace amount of H2O is indispensable. The reaction is applicable to a broad spectrum of substrates and delivers the products in good to excellent yields. Access to unsymmetrical diarylmethanes with this method is also explored and various factors are discussed.

Multi-substituted quinolines 3 were unexpectedly prepared from a cascade reaction of ortho-azido-β-nitro-styrenes with various carbonyl compounds. This method takes advantages of mild condition, simple work-up, high yield as well as wide substrate scope, which makes this method powerful for one-pot synthesis of multi-substituted quinolines. © 2013 The Authors. 摘要:
This work reports a study of protein adsorption and cell adhesion on binary self-assembled monolayers (SAMs) of alkanethiols with terminal perfluoroalkyl (PFA) and oligo(ethylene glycol) (OEG) chains in varying ratios. The surface chemistry of the SAMs was characterized by contact angle measurement, grazing angle infrared spectroscopy (GIR), X-ray photoelectron spectroscopy, and the effect on protein adsorption was investigated by surface plasmon resonance, GIR, and immunosorbent assay. Hela cell adhesion on these surfaces was also studied by fluorescence microscopy. Results reveal that, compared to OEG, PFA tended to be a higher fraction of the composition in SAM than in the assembly solution. More interestingly, the nearly 38% PFA SAM had a strong antifouling property whereas the 74% PFA SAM showed a high adsorption capacity to protein and cell. The binary PFA/OEG SAMs were favorable for maintaining the fibrinogen conformation, hence its high activity. The findings may have important implications for constructing PFA-containing surfaces with the distinct properties that is highly resistant or highly favorable toward protein adsorption and cell adhesion. This paper presents the fabrication of a molecularly imprinting sol-gel hybrid film by the one-step electrodeposition of the constitutional individuals including chitosan (CS), phenyltrimethoxysilane (PTMS), in situ formed gold nanoparticles (AuNPs) and template p-nitrophenol (p-NP). The electrodeposition was triggered by applying an optimal potential at -0.30 V vs. SCE, leading to the formation of the p-NP imprinting CS/PTMS/AuNPs hybrid film on a glassy carbon electrode (GCE) with a roughly architectural and conductive nature, as revealed by scanning electron microscopy and electrochemical impedance analysis. The mechanism of the hybrid film formation was discussed accordingly. Upon complete removal of the template molecules assisted by cyclic voltammetry, the p-NP imprinted film modified electrode exhibited differential pulse voltammetric (DPV) responses to p-NP in a linear range from 3.0 × 10 -8 to 3.5 × 10 -4 M with a detection limit of 5.0 × 10 -9 M. The selectivity and reusability of the sensor was demonstrated by discriminating the p-NP response from its analogues and successive rebinding/debinding cycles, respectively. The methodology is extendable as a simple and general platform for developing hybrid film sensors for the specific determination of various electrochemically active species. © 2013 The Royal Society of Chemistry.

Au-ZrO<inf>2</inf>-SiO<inf>2</inf> nanocomposite spheres were synthesized and used as selective sorbents for the solid-phase extraction (SPE) of organophosphorous agents. A non-enzymatic electrochemical sensor based on a Au-ZrO<inf>2</inf>-SiO<inf>2</inf> modified electrode was developed for the selective detection of organophosphorous pesticides (OPs). The Au-ZrO <inf>2</inf>-SiO<inf>2</inf> nanocomposite spheres were synthesized by the hydrolysis and condensation of zirconium n-butoxide (TBOZ) on the surface of SiO<inf>2</inf> spheres and then the introduction of gold nanoparticles on the surface. Transmission electron microscopy and X-ray photoelectron spectroscopy were performed to characterize the formation of the nanocomposite spheres. Fast extraction of OP was achieved by the Au-ZrO<inf>2</inf>-SiO<inf>2</inf> modified electrode within 5 min via the specific affinity between zirconia and the phosphoric group. The assay yields a broad concentration range of paraoxon-ethyl from 1.0 to 500 ng mL<sup>-1</sup> with a detection limit of 0.5 ng mL <sup>-1</sup>. This selective and sensitive method holds great promise for the enrichment and detection of OPs. ©2012 The Royal Society of Chemistry.

Series of 4H-3,1-benzoxazin-4-ones and 3H-quinazolin-4-ones with phenoxy-methyl substituents were rationally designed and easily synthesized via one-pot N-acylation/ring closure reactions of anthranilic acids with 2-phenoxyacetyl chlorides to yield the 4H-3,1-benzoxazin-4-ones, and subsequently substituted with amino derivatives to obtain the 3H-quinazolin-4-ones. The herbicidal evaluation was performed on the model plants barnyard grass (a monocotyledon) and rape (a dicotyledon), and most of the title compounds displayed high levels of phytotoxicity. The active substructure and inhibitory phenotype analysis indicated that these compounds could be attributed to the class of plant hormone inhibitors. A docking study of several representative compounds with the hormone receptor TIR1 revealed an appreciable conformational match in the active site, implicating these compounds are potential lead hits targeting this receptor.

We demonstrate a facile procedure to efficiently prepare Prussian blue nanocubes/reduced graphene oxide (PBNCs/rGO) nanocomposite by directly mixing Fe<sup>3+</sup> and [Fe(CN)<inf>6</inf>]<sup>3-</sup> in the presence of GO in polyethyleneimine aqueous solution, resulting in a novel acetylcholinesterase (AChE) biosensor for detection of organophosphorus pesticides (OPs). The obtained nanocomposite was characterized by X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM) and energy-dispersive X-ray (EDX) microanalysis. It was clearly observed that the nanosheet has been decorated with cubic PB nanoparticles and nearly all the nanoparticles are distributed uniformly only on the surface of the reduced GO. No isolated PB nanoparticles were observed, indicating the strong interaction between PB nanocubes and the reduced GO and the formation of PBNCs/rGO nanocomposite. The obtained PBNCs/rGO based AChE biosensor make the peak potential shift negatively to 220 mV. The over-potential decreases ∼460 mV compared to that on a bare electrode, suggesting that PBNCs/rGO has a high electrocatalytic activity towards the oxidation of thiocholine. The AChE biosensor shows rapid response and high sensitivity for detection of monocrotophos with a linear range from 1.0 to 600 ng mL<sup>-1</sup> and a detection limit of 0.1 ng mL<sup>-1</sup>. These results suggest that the PBNCs/rGO hybrids nanocomposite exhibited high electrocatalytic activity towards the oxidation of thiocholine, which lead to the sensitive detection of OP pesticides. 设计合成新型含氟单体AFA、MAFA和DAFA,分别与乙烯基三甲氧基硅烷(VTMS)和乙烯基三乙氧基硅烷(VTES)进行共聚反应得到含全氟烷基和硅氧烷基的反应性氟硅高分子。这些高分子可以与含羟基的固体表面发生硅氧化,形成固体表面修饰涂层膜,其含氟成分在分布上呈现梯度性,具有良好的疏水疏油性和稳定性。

通式为1的2-苯氧甲基-4H-3,1-苯并噁嗪-4-酮衍生物,其中,R1、R2为氢、卤素;R3为氢、卤素;R4、R5为氢、卤素、烷基、烷氧基。其合成是先将苯氧乙酸转变为苯氧乙酰氯,再与邻氨基苯甲酸在溶剂和碱存在下进行关环反应得到。用NY/T?1155.1-2006标准进行室内除草活性测试表明,这些化合物有很好的除草活性,大部分在浓度100ppm和10ppm时,表现出非选择性除草活性,除草活性高于商品氰氟草酯;在浓度1ppm时,大部分表现出选择性除草活性,与商品氰氟草酯作用相反,对单子叶植物生长抑制降低,对双子叶植物保持良好的除草活性。该类化合物作为除草剂单独使用;或者与其它植保上允许的载体或稀释剂混合配制成各种剂型使用;或与其它农药复配使用或同时并用。

This article reports the fabrication of a nanocomposite biosensor for the sensitive and specific detection of methyl parathion. The nanocomposite sensing film was prepared via the formation of gold nanoparticles on silica particles, mixing with multiwall carbon nanotubes and subsequent covalent immobilization of methyl parathion hydrolase. The composite of the individual materials was finely tuned to offer the sensing film with high specific surface area and high conductivity. A significant synergistic effect of nanocomposites on the biosensor performance was observed in biosensing methyl parathion. The square wave voltammetric responses displayed well defined peaks, linearly proportional to the concentrations of methyl parathion in the range from 0.001μgmL<sup>-1</sup> to 5.0μgmL<sup>-1</sup> with a detection limit of 0.3ngmL<sup>-1</sup>. The application of this biosensor in the analysis of spiked garlic samples was also evaluated. The proposed protocol can be used as a platform for the simple and fast construction of biosensors with good performance for the determination of enzyme-specific electroactive species. ©2011 Elsevier B.V.

本发明公开了有机磷农药生物标志物的检测方法。该方法利用肟类复活剂对抑制后乙酰胆碱酯酶(AChE)的复活作用,用线性扫描法分别测量复活前、后AChE的活性,根据两者之间的活性差异,得到对应乙酰胆碱酯酶加合物OP-AChE含量,从而判断农药的接触水平。该方法以复活后AChE含量作为个体自身的基准,消除了个体间正常水平AChE的变异性。
