Effect of combined fish meal : Soybean meal ratio, vitamin C and fish oil supplementations in diet on the growth and reproduction of red swamp crayfish, Procambarus clarkii (Crustacea: Decapoda)

摘要:
Fish meal (FM), vitamin C (VC) and fish oil (FO) play important roles in the growth and reproduction in aquatic animals. To determine the optimum dietary combinations of FM:SM (soybean meal) ratio, VC and FO supplementations for the growth and reproduction of female crayfish Procambarus clarkii, we used the L9 (33) orthogonal array, which arranged three factors varied at three levels (FM:SM at 1:3, 1:2 and 1:1.5; VC supplementation at 0.02%, 0.04% and 0.06%; FO inclusion at 1.90%, 2.85% and 3.80%). Range analysis showed that the highest weight gain (WG) of the crayfish occurred at FM:SM of 1:1.5, VC supplementation of 0.04% and FO inclusion of 1.90%, and the maximum gonadosomatic index (GSI), egg laying amount (ELA) and spawning rate (SR) were obtained at FM:SM 1:1.5, VC supplementation 0.06% and FO inclusion 3.80%. Analysis of variance indicated that all the three factors had no significant effects on WG. Fish oil supplementation significantly affected GSI, ELA and SR (P<0.05), while FM:SM did not significantly influence any of these indicators. Taking the cost into consideration, we suggest that a dietary FM:SM of 1:3, VC supplementation 0.06% and FO inclusion 3.80% might be the optimal combinations for the reproduction of P. clarkii.

Hira基因产物在银鲫和彩鲫卵子发生过程中的动态变化

摘要:
为进一步研究Hira基因在卵子发生和雌核发育过程中的作用,通过原位杂交和免疫荧光定位的方法 检测了Hira mRNA和蛋白质在雌核发育银鲫和两性生殖彩鲫卵子发生过程中的动态变化.结果表明,银鲫 和彩鲫卵子发生过程中tara基因转录产物的变化基本一致,在I期卵母细胞的细胞核中大量表达,至Ⅱ期 卵母细胞时转至细胞质中均匀分布,在Ⅲ期卵母细胞中,杂交信号逐渐移向细胞的周边,到Ⅳ期时随着卵黄 物质大量积累,杂交信号几乎不见.HIRA蛋白在银鲫和彩鲫卵子发生过程中的变化略有差别.HIRA蛋白 在银鲫I期卵母细胞中没有表达,在Ⅱ期卵母细胞的细胞质中有弱表达,在Ⅲ期早期卵母细胞的周边有强 烈表达;而在彩鲫I期卵母细胞中就有HIRA蛋白的弱表达,至Ⅱ期时HIRA蛋自在细胞质中大量表达,Ⅲ 期早期卵母细胞的细胞质中有弱表达.在银鲫和彩鲫Ⅲ期末和Ⅳ期卵母细胞中都很难观察到荧光信号.Hira mRNA和蛋白质在银鲫和彩鲫早期卵母细胞中有较强表达,且在银鲫和彩鲫卵子发生过程中没有显著差异, 说明其可能对于脊椎动物卵子发生和减数分裂没有显著影响,而是在受精和/或胚胎发育过程中起作用.

随着时代的进步和高等教育的发展,构建开放式、研究型的课堂已成为高校教育发展的趋势。针对当前高校生物科学专业教学形式单一,忽略学生主体性的发挥,以及评价方式不能客观考核学生操作技能等现状。文章以行为主义、建构主义教育理论为指导,将高校生物科学专业分为基础性实验、研究性实验和自主设计性实验三类,并分别构建其教学模式和评价体系,以期为当前的实践教学改革提供指导。

摘要:
The red swamp crayfish, Procambarus clarkii (Girard), has become an important freshwater species for Chinese inland aquaculture because of its high commercial value and consumer demand. The aim of this study was to gather information about the activity of digestive enzymes at different embryonic stages of P. clarkii in order to increase our knowledge about digestive physiology and to guide technology for maternal culture so as to improve the hatching rate. Embryonic developmental stages were divided into six stages: I, fertilized egg; II, cleavage and blastula; III, gastrula; IV, egg nauplius; V, eye pigment forming; and VI, prepared for hatching. Pepsin-specific activity decreased significantly from stage I to stage IV. Although it increased at stage V, the activity level declined again before hatching. Both trypsin- and amylase-specific activity levels dramatically increased in the middle stages of embryogenesis, whereas at the other embryonic stages the activities of these two digestive enzymes were much lower. Lipase-specific activity exhibited a low level during all embryonic stages. The pattern of digestive enzyme activity was related to organogenesis and the utilization of yolk proteins at different embryonic stages. © 2009 Blackwell Publishing Ltd.

The red swamp crayfish, Procambarus clarkii (Girard), is one of the most commonly farmed freshwater species in inland China due to its high market value and consumer demand. The aim of this study was to determine the optimum combinations of photoperiod, water calcium concentration and pH for juvenile survival, growth and moulting. In our orthogonal experiment, the three environmental factors were varied at three levels (photoperiod: 16L:8D, 12L:12D and 8L:16D; calcium concentration: 45.5, 65.5 and 85.5 mg L-1; and pH: 6.8, 7.8 and 8.8). Range analysis showed that the maximum survival of juvenile crayfish occurred at photoperiods of 16L:8D or 8L:16D, water calcium concentration of 45.5 mg L-1 and pH of 7.8; maximum weight gain at photoperiod 16L:8D, water calcium concentration 65.5 mg L-1 and pH 7.8; maximum length increase at photoperiod 16L:8D, water calcium concentration 65.5 mg L-1 and pH 7.8; and the highest moult frequency at photoperiod 12L:12D, water calcium concentration 65.5 mg L-1 and pH 7.8. Analysis of variance indicated that photoperiod, water calcium concentration and pH significantly influenced only the weight gain of juvenile crayfish (P<0.05). Taking growth into consideration, we suggest that a photoperiod of 16L:8D, calcium concentration of 65.5 mg L-1 and pH 7.8 might be optimal conditions for rearing juvenile P. clarkii. © 2009 Blackwell Publishing Ltd.

The endosymbiotic bacterium Wolbachia infects various insects and is primarily known for its ability to manipulate host reproduction. Recent investigations reveal that Wolbachia also affects the activity of somatic cells. We here demonstrated by trap method and T-maze that Wolbachia infection had significant impact on the olfactory response of Drosophila simulans. Wolbachia-infected flies took shorter time to enter the food trap and were more sensitive to odorant in T-maze than those uninfected controls. The time of olfactory response was relative to Wolbachia density in flies. Wolbachia density in 15-day-old flies that were caught in a shorter time (less than 60 min) by food trap was significantly higher than those taken in a longer time (more than 100 min). Quantitative RT-PCR showed that the transcript of an important odorant receptor gene or83b in flies with fast olfactory response was significantly more than those with slow olfactory response. These results suggest that Wolbachia might increase olfactory response of flies by regulating the expression of olfaction-related genes in hosts.

芳香化酶(P450arom)是雌激素合成过程中的关键酶,在性别分化中起重要作用.鱼类存在卵巢性和脑型两种芳香化酶,分别由Cyp19a和Cyp19b编码.稀有鮈鲫作为我国特有的实验动物,尚无其芳香化酶的有关资料,其性别分化机制亦不清楚.本研究采用RT-PCR的方法以简并引物扩增了稀有鮈鲫脑型芳香化酶基因Cyp19b的部分序列,其长度为1070 bp,编码357个氨基酸残基,具有典型的芳香化酶结构域.RT-PCR分析发现该基因主要在稀有鮈鲫的脑中表达,在性腺、肠、肾中也有表达;其在雌、雄脑中的表达差异不显著.在胚胎发育阶段,Cyp19b的表达从囊胚期开始,至神经胚期达到较高水平,随后下降,孵化期又有所增强,孵化10d后维持在高水平.这些结果说明Cyp19b以脑中表达为主,可能在稀有鮈鲫神经系统发育和脑的性别分化中有重要作用.

摘要:
Most pathogens require a relatively long period of development in their mosquito vector before they can be transmitted to a new human host; hence, only older insects are of epidemiological importance. The successful transfer of a life-shortening strain of the inherited bacterial symbiont, Wolbachia, into the major mosquito vector of dengue, Aedes aegypti, halved adult life span under laboratory conditions. The association is stable, and the Wolbachia strain is maternally inherited at high frequency. It is capable of inducing complete cytoplasmic incompatibility, which should facilitate its invasion into natural field populations and its persistence over time. Our data suggest that targeting mosquito age with inherited Wolbachia infections may be a viable strategy to reduce the transmission of pathogens such as dengue viruses.

采用正交设计法,探讨了水温、光周期和饲料组成3个因素对克氏原螯虾雌虾成活和性腺发育的影响,以期为人工繁殖提供理论指导.结果表明,各因素对克氏原螯虾雌虾成活率的影响大小为:水温饲料组成光周期,其最优水平分别为:水温22℃、光照周期16L: 8D、饲料为纯草鱼肉.以性腺成熟系数为指标,3个因素对雌虾性腺发育的影响大小为:饲料组成水温光周期,其最优水平分别为:饲料为纯草鱼肉、水温28℃、光照周期16L : 8D.根据本实验结果,综合考虑成活率和性腺发育情况,克氏原螯虾的亲虾培育可选择水温25~28℃之间、光照周期16L : 8D.饲料中多添加鱼肉等动物性饲料.同时还应注意减小饲养密度,以免发生同类残食的事件.

一种简便的适合抱卵孵化甲壳动物的转基因方法

动物转基因的方法有很多,但对于许多抱卵孵化的甲壳动物,许多已有的转基因方法不可行或不易操作。在对罗氏沼虾(Macrobrachium rosenbergii)的基因改良过程中,通过精荚微注射(spermatophore-microinjection, SMI),将目的基因直接注射到交配后的雌虾胸部腹面的精荚中,再由精子介导的简便高效的转基因技术,成功地将抗冻蛋白基因导入到罗氏沼虾的胚胎中,获得了16%以上的基因转移率。这一方法的建立,为进一步开展抱卵孵化的甲壳动物的分子遗传和发育生物学的研究以及育种工作的研究奠定了基础。

HmgD encodes Drosophila homologue of high mobility group proteins (HMGD), which are thought to have an architectural function in chromatin organization. However, current opinions about the function of HMGD in Drosophila development ore controversial. Our previous studies have shown that ubiquitous overexpression of HmgD caused the formation of melanotic tumors in the Drosophila larvae by prematurely activating the Ras-MAPK pathway. Here we report that under maternal control, the viability of flies links with overexpression of HmgD, while under ubiquitous control, ActGal4, overexpressing HmgD animals, which display prolonged larval stages around day 13, developmentally stagnate in the larva-white pupa transition. Ecdysone feeding did not rescue overexpressing HmgD animals. RT-PCR analyses show that overexpression of HmgD does not affect the temporal expression pattern of ecdysone receptor gene EcR, whereas transcriptional patterns of some key regulatory genes, such as E74A, E74B, E75A, E75B, βETZ-F1, are changed greatly. These results suggest that ubiquitous overexpression of HmgD results in the failure of pupariation neither by affecting the process of ecdysone synthesis ond release nor by abnormal EcR transcription, but by causing expression of EcR regulatory nuclear receptors out of schedule. The results led us to postulate that overexpression of HMGD likely changes the signaling cascade of Drosophila metamorphosis by an interaction between HMGD and DNA strands, and subsequently by on error of DNA binding abilities and transcriptional activities of some nuclear receptor genes. © 2008 Wiley-Liss, Inc.

Hir/Hira基因家族的成员广泛存在于多种生物体中,但有关其在生物体发育过程中的具体功能还不甚清楚。对果蝇的研究表明,dHira基因产物可能在受精时精核的解凝过程和雄性原核的正常形成过程中起重要作用。本研究组前期已经分别克隆出雌核发育银鲫和两性生殖彩鲫的Hira基因(cagHira和caHira),本实验在银鲫cagHira基因的特异区域,设计一对引物,以银鲫成熟卵母细胞总RNA逆转录出的cDNA为模板,扩增出cagHira的特异片段。再将该片段克隆到原核表达载体pET-32a上,转化BL21(DE3)菌株,经诱导后表达出融合蛋白。分析表明,该融合蛋白主要以包涵体形式表达。以纯化的融合蛋白作为抗原去免疫小鼠,制备多克隆抗血清,经蛋白质印迹分析检测,该抗血清(稀释到1∶2000)与包涵体蛋白识别反应良好,确定获得了具有高效价的特异性银鲫CAGHIRA多克隆抗体,为进一步研究HIRA在鱼类发育和雌核生殖过程中的作用奠定了基础。对银鲫HIRA蛋白的组织特异性表达分析发现,该蛋白仅在成熟卵巢组织中特异表达,故表明HIRA可能对鱼类卵子发生和/或早期胚胎发育具有重要作用。

武汉地区克氏原螯虾繁殖期的研究

摘要:
为探究武汉地区克氏原螯虾(Procambarus clarkii)繁殖期的问题,采用石蜡切片的方法,对武汉地区自然群体中克氏原螯虾雌雄成体的生殖腺进行了周年观察研究.结果表明,武汉地区成年克氏原螯虾的卵母细胞发育不同步,在4~5月以及9~10月的卵巢中都有大量成熟期的卵子,同时也含有少量卵黄发生前期或初期的卵母细胞.6~7月和11月都可见产后恢复期的卵巢,内含少量未产出的正在退化的过熟卵和一些卵黄发生前期或初期的卵母细胞.精巢发育的年周期变化没有卵巢变化的那么明显,但也可以看到,在4~6月和9~10月,含精子细胞和精子的精小管的比例大大增加,而在其他时期,含精原细胞和精母细胞的精小管比例较多.这些结果提示,武汉地区克氏原螯虾一年有两个繁殖高峰期.一个在4~5月,另一个在9~10月.
