摘要:
Cobalt ferrite (CoFe2O4) nanocrystals (approximately 10 nm) were prepared by hydrothermal method and their interactions with Bovine Serum Albumin (BSA) and bovine hemoglobin were investigated by Zeta potential, Dynamic Light Scattering (DLS) and FTIR Spectroscopy techniques. Results show that nanocrystal-hemoglobin adsorption correlates to electrostatic attractive/repulsive interactionB whereas BSA does not follow this scheme. The corresponding adsorption capacity of BSA and hemoglobin reaches a maximum value of 237.9 mg · g−1 and 256.9 mg · g−1 at pH value of 5.5 and 7.0, respectively. DLS measurements indicate that protein adsorption have led to nanocrystal-protein aggregates formation. Comparing to the hydrodynamic size of bare nanocrystals (51 nm), that for BSA and hemoglobin increases to 472 nm and 114 nm respectively upon protein adsorption. The interaction with nanocrystal also induces protein conformation changes. The amide I band in IR spectrum shifts 4 cm−1 and 6 cm−1, respectively, to lower wavenumbers in the case of BSA and hemoglobin. © 2014, Chinese Chemical Society. All rights reserved.

摘要:
[目的]分离得到高效的邻苯二甲酸二乙基己基酯(DEHP)降解菌。[方法]采用富集培养法筛选分离菌株,并对菌株进行驯化;通过PCR 扩增得到其16S rRNA 和gyrB 基因序列,进行同源序列分析及分子系统发育树的构建,同时结合形态学观察和生理生化实验对菌株进行初步鉴定;采用高效液相色谱(HPLC)分析菌株对DEHP 的降解特性。[结果]分离得到一株能以DEHP 为唯一碳源和能源生长的菌株,命名为HS-NH1,初步鉴定其为戈登氏菌(Gordonia sp.)。菌株HS-NH1最适的生长和降解条件为30 °C、pH 7.0,在此条件下,该菌株60 h 内能够将浓度为500 mg/L 的DEHP 降解90%以上。高效液相色谱(HPLC)分析表明,菌株HS-NH1在降解DEHP 过程中产生了一种重要的中间代谢产物——邻苯二甲酸。底物广谱性试验证明,菌株HS-NH1能够有效地利用多种常见的邻苯二甲酸酯(PAEs)与芳香族衍生物。[结论]筛选得到了一株DEHP 降解菌Gordonia sp. HS-NH1,该菌降解效率高,具有良好的底物广谱性,在邻苯二甲酸酯类化合物的污染治理中将会有一定的应用潜力。

摘要:
In this study, a bacterium strain, CZ-31, capable of degrading methyl parathion (MP) was isolated from active sludge of a wastewater treatment pond of a pesticide factory in Hubei province. According to morphological characteristics, physiological properties, and 16S rDNA sequence analysis, the isolates CZ-31 were identified as Arthrobacter ureafaciens. Methyl parathion degradation enzyme mpd gene was cloned by PCR from the genome of strain CZ-31. Nucleotide sequencing indicated that the mpd gene contains 996bp and is predicated to encode a peptide of 331 amino acids. The recombinant expression plasmid pET-mpd was constructed and successfully expressed in E. coli BL21 (DE3). The structure of methyl parathion degradation enzyme (MPD) was predicted by bioinformatics. Results showed MPD protein was 97.28% similar to MPH protein (from pseudomonas sp. WBC-3) and it belonged to metallo-beta-lactamases family. The model consists of 588 residues, three Zn2+, and one Cd2+. There is a homodimer comprising of two independent subunits in MPD structure. Each subunit is composed of alpha beta-lactamase-like domain, which includes the binuclear metal center. The binuclear metal site is located between the two beta-sheets and is surrounded by two alpha beta-loops. Homology modeling of MPD was constructed based on the crystal structure of MPH (PDBID:1P9E).

摘要:
In order to explore the impact of mercury on fish, zebrafish were exposed in various levels of HgCl2 and the superoxide dismutase (SOD), acetylcholinesterase (AChE) activities were measured after 1 and 7 days exposure. Muscle RNA was also extracted for semi-quantitative RT-PCR determination of the expression of AChE, CYP1A1, Cu/Zn-SOD. It indicated that 96 h-LC50 of HgCl2 to zebrafish is 244.06 µg L−1. The AChE activity was decreased on 1st day and 7th day. The SOD activity was almost unchanged on 1st day, it showed a dose–response effect on 7th day. The expression of AChE was down-regulated on 1st day, it is consistent with the AChE activity, but the expression was showed a significant difference between exposed group and the control group on 7th day. The expression of CYP1A1 was decreased compared with the control group on 1st day. After 7 day exposure, the expression of CYP1A1 was up-regulated in 0.1LC50 group, and down-regulated in 0.4LC50 group, 0.8LC50 group, showing a stimulation effect at low concentration and inhibition at high concentration. The expression of Cu/Zn-SOD was down-regulated on 1st day, also showing an effect of stimulation at low concentration and inhibition at high concentrations. Thus HgCl2 was highly toxic to zebrafish, both the enzymatic activities and the gene transcription were changed significantly. But the change of gene transcription is earlier than enzymes activities. It indicates that gene transcript can be used as an early warning for environmental risk assessment. © 2014, Springer Science+Business Media New York.

摘要:
In order to observe the toxic effects of butyl benzyl phthalate (BBP) on zebrafish, the AChE and SOD activity of zebrafish exposed to different concentrations of BBP (0, 0.332, 0.665, 1.33 mg L −1 ) in a short-term (7d) test were determined. Semi-quantitative PCR was used to determine the mRNA transcript levels of the AChE and SOD gene in zebrafish brain and muscle. The results showed: AChE activity decreased with increased exposure concentration, and was significantly inhibited (p < 0.01) compared with the control group at 0.665 mg L −1 concentration. Low BBP concentrations stimulated and high concentrations inhibited SOD activity with a concentration of 0.332 mg L −1 resulting in a significant induction (p < 0.05) compared with the control, and 0.665 and 1.33 mg L −1 concentrations resulting in significant inhibition (p < 0.05, p < 0.01) relative to the control group. The RT-PCR data showed a decrease in brain and muscle mRNA transcription of AChE gene with an increase in exposure concentration. The mRNA transcription of SOD in the brain was not different between the exposed groups and control group; in muscle, the mRNA transcription inhibition decreased and then increased: all differences from the control were statistically significant. © 2014, Springer Science+Business Media New York.

摘要:
Green mold caused by Penicillium digitatum is one of the most serious postharvest diseases of citrus fruit, and it is ubiquitous in all citrus growing regions in the world. Sterol 14α-demethylase (CYP51) is one of the key enzymes of sterol biosynthesis in the biological kingdom and a prime target of antifungal drugs. Mutations in CYP51s have been found to be correlated with resistance to azole fungicides in many fungal species. To investigate the mechanism of resistance to prochloraz (PRC) in P. digitatum, the PRC sensitivity was determined in vitro in this study to assess the sensitivity of 78 P. digitatum isolates collected in Hubei province. The results showed that 25 isolates were prochloraz-resistant (PRC-R), including six high-resistant (HR) strains, twelve medium-resistant (MR) and seven low-resistant (LR) strains. A sequence analysis showed no consistent point mutations of PdCYP51A in the PRC-R strains, but four substitutions of CYP51B were found, Q309H in LR strains, Y136H and Q309H in HR strains, and G459S and F506I in MR strains, which corresponded to the four sensitivity levels. Based on the sequence alignment analysis and homology modeling followed by the molecular docking of the PdCYP51B protein, the potential correlation between the mutations and PRC resistance is proposed.

摘要:
Nitrate is considered to be one of the most widely present pollutants leading to eutrophication of environment. The purpose of this work was to isolate and identify new aerobic denitrifying bacteria from nitrate-contaminated sediments and utilize modified immobilized techniques for immobilizing isolates to improve nitrate removal efficiency. Using traditional enrichment approach, three purified aerobic bacteria (HS-N2, HS-N25 and HS-N62) capable of NO<inf>3</inf><sup>-</sup>-N removal from sediments were obtained. According to their physiological properties and 16S rRNA gene sequences analysis, isolates HS-N2, HS-N25 and HS-N62 were identified as Salmonella sp., Bacillus cereus and Pseudomonas sp. respectively. After cultivating at 37 °C with 150 rpm shaking, the percentages of NO<inf>3</inf><sup>-</sup>-N removal by isolates HS-N2, HS-N25 and HS-N62 were 95% in 24 h, 100% in 24 h and 96% in 12 h, respectively. Due to its higher removal rate of NO<inf>3</inf><sup>-</sup>-N, HS-N62 was selected for immobilization with polyvinyl alcohol (PVA)-sodium alginate (SA) and powder activated carbon. The removal efficiency of NO<inf>3</inf><sup>-</sup>-N by immobilized HS-N62 pellets was enhanced with no nitrite accumulation. Immobilized pellets exhibited more desirable denitrification after 24 h under stressful batch conditions, indicating they had stable and strong denitrifying capabilities. ©2012 Elsevier Ltd.

摘要:
nButyl benzyl phthalate (BBP) is a phthalic acid ester (PAE) used extensively in the manufacturing of plastics that contributes to the environmental pollution. It easily released to the environment and resistant to degradation. Bioremediation has been proposed for removal of PAEs from contaminated sites. Studies showed that the biodegradation of PAEs was slow if the lateral chain was longer and more complex. Two BBP degrading bacterial strains, HS-B1, HS-B2, were isolated from contaminated sludge in the river of Jingzhou in Hubei province. The two bacteria were identified as Acinetobacter sp. and Arthrobacter sp. by morphology and 16S rDNA sequence analysis. BBP at 500 mg L<sup>-1</sup> in the culture medium was tested and after 24 h of culture, chloroform was used to extract the BBP in the culture medium. The initial and the final concentrations of BBP were assessed by UV-visible spectrophotometer. The results showed that the bacterium could grow well on BBP as the sole source of carbon and energy. The degradation of 500 mg L<sup>-1</sup> BBP by strains HS-B1 and HS-B2 was 28% and 59% after 24 h, respectively. The rate of degradation increased to 40%, 75% respectively with addition of 0.1% LB in 24 h, suggesting that additional sources of growth factors enhanced biodegradation. This study is helpful to the development of microbial resources for bioremediation. ©2012 Elsevier Ltd.

Objective Cartilage oligomeric matrix protein (COMP) is a major noncollagenous component of cartilage and is used as a biomarker in rheumatoid arthritis and experimental arthritis. Injection of COMP leads to severe inflammatory joint disease, and antibodies play a critical role in mediating arthritis. The arthritogenicity of COMP might be due to the lack of self tolerance. This study was undertaken to determine the status of COMP-specific B cell tolerance using COMP-deficient mice. Methods Arthritis development and antibody responses were compared between COMP-sufficient and COMP-deficient littermates after immunization with rat COMP. Serum anti-COMP antibody levels were measured using a panel of recombinant mouse COMP proteins, and antibody-secreting cells were enumerated by enzyme-linked immunospot assays. A novel sandwich enzyme-linked immunosorbent assay was developed to assess COMP molecules in serum. Results COMP-sufficient mice, but not COMP-deficient mice, developed severe arthritis following immunization with rat COMP. However, anti-COMP antibody titers to native COMP and recombinant protein domains covering the entire mouse COMP sequence, except the less immunodominant type 3 repeat domains, were decreased in COMP-sufficient mice compared to COMP-deficient mice. In addition, COMP-sufficient mice had fewer B cells secreting COMP-reactive antibodies. Detectable levels of full-length COMP in arthritic COMP-sufficient B10.Q NCF-1*/* and healthy mice suggested systemic availability of COMP to the immune system. Conclusion The lack of arthritis, together with high levels of COMP-specific antibodies, in COMP-deficient mice indicates that susceptibility to arthritis is COMP specific and that endogenous expression of COMP in wild-type mice tolerizes B cells in vivo. 通过提高鱼体自身免疫力的免疫预防是防控罗非鱼链球菌病的重要对策,其中新型高效疫苗的研制将是罗非鱼链球菌病预防的主要发展方向。目前在研的罗非鱼链球菌病疫苗有灭活疫苗、减毒疫苗、亚单位疫苗、DNA疫苗、被动免疫疫苗等多种形式。而疫苗的接种途径、佐剂、鱼体状态和环境因素等对疫苗的效果有重要影响。文章对罗非鱼链球菌病疫苗的研究现状做一综述,以期为该疫苗的设计和应用提供科学对策。

目的:为了揭示循环血液中存在的软骨寡聚基质蛋白(Cartilage Oligomeric Matrix Protein,COMP)片段,探索COMP逃避自身免疫耐受的途径及机制,需制备高效、专一的识别COMP不同结构域的单克隆抗体。方法:利用真核表达系统对人COMP进行了一系列的表达、并用亲和层析法结合离子交换层析法进行纯化。纯化的COMP为抗原免疫BALB/c小鼠,采用杂交瘤技术结合有限稀释传代法筛选得到稳定分泌抗人COMP单克隆抗体的杂交瘤细胞株,用COMP单结构域片段结合生物信息学方法分析鉴定单克隆抗体结合表位。用不同种属来源的COMP分析单克隆抗体结合表位的保守性。结果:COMP及单结构域片段为分泌到细胞外的可溶性分子利于蛋白的纯化,筛选获得4株稳定分泌抗人COMP单克隆抗体细胞株,4株单克隆抗体的亚型为IgG1,经COMP单结构域片段检测证明4株单克隆抗体识别不同的表位,表位预测软件初步确定4株单克隆抗体识别表位所在区域,间接ELISA及免疫组化证实其中3株单克隆抗体与不同种属来源的COMP有较好的结合特性。结论:成功制备了4株识别不同表位的抗COMP单克隆抗体,为揭示血液中存在的COMP片段和进一步研究COMP逃避自身免疫耐受机理提供了研究工具。

纳米硫化镉量子点对斜生栅藻的毒性研究

摘要:
采用96 h急性毒性实验的方法,研究了新型纳米材料硫化镉量子点(CdS)对斜生栅藻的毒性效应。结果表明,随着量子点浓度的增大,其对斜生栅藻生长的抑制作用也在增大,并在浓度为1.0 mg·L~(-1)时达到了显著抑制作用。通过光学显微镜观察发现,经过量子点处理之后的斜生栅藻细胞形态出现了变化,并且随着处理浓度的增大,藻细胞颜色变浅的程度也增大,这说明硫化镉量子点可能对藻细胞色素的合成存在阻碍作用。在处理96 h后,硫化镉量子点对藻细胞光合色素含量的影响和藻密度的影响是一致的,各浓度组SOD(超氧化物歧化酶)含量相比于对照组均出现了显著变化,而MDA(丙二醛)含量则是在浓度为0.2、0.4、0.6 mg·L~(-1)时与对照组相比变化较小,在浓度为0.8、1.0 mg·L~(-1)时出现较显著变化,并且SOD 和MDA含量在浓度为1.0 mg·L~(-1)时呈明显负相关关系,这说明在各组均发生了不同程度的脂质过氧化反应。

1株高效BBP降解菌的分离与特性研究

摘要:
从湖北省荆沙河泥样中富集培养分离得到了1株能够以邻苯二甲酸丁基苄基酯为唯一碳源和能源生长的细菌并命名为HS-B1.采用形态学、生理生化和16S rDNA序列分析对其进行了鉴定,发现HS-B1是1株杆状、需氧、革兰氏阴性细菌,接触酶呈阳性反应,氧化酶呈阴性反应,不产硫化氢气体,且该菌的16S rDNA序列与琼氏不动杆菌(Acinetobacter junii)的相似性高达99%.因此初步鉴定该菌株为不动杆菌(Acinetobacter sp.).摇瓶实验表明,菌株HS-B1生长和BBP降解的最适培养条件为35℃,pH 8.0.采用0.10 mmol·L~(-1)非离子型表面活性剂吐温-80提高了BBP在水溶液中的表观溶解度,发现在最适培养条件下,HS-B1能够在48 h内将浓度为1 000 mg·L~(-1)BBP完全降解,证明是一株高效降解菌.底物广谱性实验中,菌株HS-B1也能够有效利用DMP、DEP、DBP等邻苯二甲酸酯类化合物,说明该菌株在处理邻苯二甲酸酯类化合物的污染治理中有独特的应用潜力.

摘要:
铁氧还蛋白还原酶(FerredoXin-NADP~+reductases, FNR)是一种在生物体的多种代谢中起着重要作用的黄素酶.为探讨嗜水气单胞菌(Aeromonas hydrophila)XS91-4-1中FNR的功能和结构,通过克隆其FNR基因,构建重组表达载体pET42a-fnr,表达并分离纯化了具有活性的GST-FNR融合蛋白,根据米曼氏动力学测定了纯化蛋白对NADPH及EDTA-Fe~(3+)的酶活力,采用生物信息学方法对XS91-4-1 FNR蛋白序列进行系统学分析,并初步预测了该蛋白三维结构. 结果表明:重组质粒能够在宿主菌大肠杆菌(Escherichia coli) BL21中获得高效可溶性表达,经镍柱亲和层析法可纯化出电泳均一的目的蛋白,蛋白浓度为67.3 μg/mL;纯化蛋白对NADPH的比活力为1.78 U/mg,对EDTA-Fe~(3+)的比活力为1.13 U/mg,比活力比粗酶分别提高了约29和22倍;生物信息学分析表明XS91-4-1 FNR分布在Bacterial-class FNR分支上且具有与此类FNR相似的结构特点.上述结果显示嗜水气单胞菌FNR具有与已报道的FNR相似的基本特性,并推测其可能属于Bacterial-class FNR.图6表1参22

摘要:
Blue mold, caused by Penicillium. italicum, is one of the most damaging postharvest diseases of citrus fruit. P. italicum Sterol 14α-demethylase (PiCYP51), an important enzyme in membrance sterol biosynthesis, is a key target of antifungal compounds for citrus disease caused by P. italicum. The three-dimensional structure of PiCYP51 from P. italicum Chinese isolate (HS-1) was constructed through homology modeling basing on the crystal structure of human CYP51. After molecular dynamics (MD) simulation, the refined model was assessed by PROCHECK on the quality. Following evaluation on the reliability was performed by investigating the binding interaction of two commercial sterol 14α-demethylase inhibitors (DMIs) with the enzyme. The binding mode predicted by the molecular docking revealed that the DMIs interacted with PiCYP51 mainly through hydrogen-bonding and hydrophobic interactions. Furthermore, the results were compatible with the detected EC50 values, which were determined as 0.25 and 0.31mg/L for tebuconazole and diniconazole. The binding mode of antifungal agents with PiCYP51 can provide references for DMIs optimization, virtual screening, or de novo antifungal compounds design.

摘要:
水热法制备了超细ZnFe_2O_4纳米晶,并通过高分辨透射电镜(HRTEM)、XRD和EDX技术对其进行了表征。以牛血清白蛋白(Bovine Serum Albumin,BSA)和牛血红蛋白(Hemoglobin)为模式蛋白,研究了纳米晶在不同pH条件下对蛋白质分子的吸附及其与ζ电势的关系。通过动态光散射(Dynamic Light Scattering,DLS)和傅立叶变换红外光谱(Fourier Transform Infrared,FTIR)技术分别研究了纳米晶吸附蛋白后流体力学直径的变化以及蛋白质构象的变化。结果表明,ZnFe_2O_4纳米晶对牛血红蛋白的吸附符合静电吸附的规律,而对BSA的吸附则不符合静电吸附的规律。纳米晶吸附牛血红蛋白后主要以单体和三聚体的形式存在,仅存在少量的团聚体,而吸附BSA后完全以团聚体的形式存在。FTIR光谱则显示纳米晶对牛血红蛋白构象的影响大于对BSA构象的影响。在合适的pH条件下,纳米晶对BSA及牛血红蛋白的吸附容量均超过380mg·g~(-1),有望应用于蛋白质的高效分离。

摘要:
[目的]探究呋喃丹和氯氰菊酯的类雌激素活性。[方法]采用重组人雌激素受体α基因的双杂交酵母测定了17β-雌二醇和2种农药诱导产生β-半乳糖苷酶活性的EC50值。[结果]17β-雌二醇、呋喃丹、氯氰菊酯对应的EC50值分别为4.04×10-10、2.34×10-7、1.04×10-6mol/L,诱导的最大酶活分别为4.97、1.82、1.59 U。[结论]2种农药均具有不同程度的类雌激素活性,且呋喃丹的类雌激素活性高于氯氰菊酯。

研究背景:软骨寡聚基质蛋白(Cartilage Oligomeric Matrix Protein,COMP)是是关节软骨的主要构成成分,占关节软骨湿重的1%。在骨关节病变早期,COMP被释放到关节腔、血液中。迟发性 RA病人关节症状尚未表现时,血清中COMP水平已经有所升高,。因此有必要建立敏感、特异的方法用于检测血液及关节滑液中COMP水平的变化。而单克隆抗体具有高度均质性和高度特异性的特点,在生物医学领域得到广泛的应用。本研究旨在建立一种敏感、特异的方法来进行COMP的检测,并为进一步研究COMP在骨关节炎的诊断中的作用其及其功能分析等奠定基础。方法:采用淋巴细胞杂交瘤技术,获得2株能稳定分泌抗COMP单克隆抗体的杂交瘤细胞株1D9、和3F12,采用间接ELISA法测定2株单抗的生物学性质。结果:2株单抗的亚类均为Ig G1,间接ELISA法测定其腹水效价分别为3×107、2×106,亲和常数分别为8.65×105 L/Mol和1.24×106 L/Mol。证明产生的2株McAb特异性好,亲和力高。结论:本研究制备了2株COMP蛋白的单克隆抗体,旨在建立一种敏感、特异的方法来进行COMP的检测,并为进一步研究COMP在骨关节炎的诊断中的作用其及其功能分析等奠定基础。
