A water-soluble fluorescent sensor, 1, based on the quinoline platform, demonstrates femtomolar sensitivity for zinc ion with a 14-fold enhanced quantum yield upon chelation to zinc ion and also exhibits high selectivity to zinc ion over other physiological relevant divalent metals in the presence of EDTA. X-ray crystal structure of zinc complex reveals that an acetic carboxylic group participates in coordination, which significantly enhances the affinity of 1 for zinc ion.

摘要:
Observations under an environmental scanning electron microscope showed that the peritrophic membrane (PM) of Spodoptera exigua (Hübner) was smooth without pores, but there were many pores and slits in the PM of larvae fed with 10 g L−1 optical brightener Calcofluor white M2R. After incubation of S. exigua PM in vitro with 10 g L−1 M2R, a significant amount of proteins was released from the PM structure. M2R disrupted the structure of larval PM, resulting in greatly increased permeability and increased larval susceptibility to Syngrapha falcifera multiple nucleopolyhedrovirus infection. Continuous feeding of larvae on a diet treated with a 10 g L−1 M2R solution significantly retarded larval development and resulted in high mortality. The destructive effect of M2R on PM was transient and reversible, depending on the presence of M2R within the midgut. Copyright © 2007 Society of Chemical Industry

3-Hydroxy-3-methylglutaryl-coenzyme A reductase (HMGR) catalyzes the formation of mevalonate. In many classes of organisms, this is the committed step leading to the synthesis of essential compounds, such as cholesterol. However, a high level of cholesterol is an important risk factor for coronary heart disease, for which an effective clinical treatment is to block HMGR using inhibitors like statins. Recently the structures of catalytic portion of human HMGR complexed with six different statins have been determined by a delicate crystallography study (Istvan and Deisenhofer Science 2001, 292, 1160-1164), which established a solid basis of structure and mechanism for the rational design, optimization, and development of even better HMGR inhibitors. In this study, three-dimensional quantitative structure-activity relationship (3D QSAR) with comparative molecular field analysis (CoMFA) was performed on a training set of up to 35 statins and statin-like compounds. Predictive models were established by using two different ways: (1) Models-fit, obtained by SYBYL conventional fit-atom molecular alignment rule, has cross-validated coefficients (q(2)) up to 0.652 and regression coefficients (r(2)) up to 0.977. (2) Models-dock, obtained by FlexE by docking compounds into the HMGR active site, has cross-validated coefficients (q(2)) up to 0.731 and regression coefficients (r(2)) up to 0.947. These models were further validated by an external testing set of 12 statins and statin-like compounds. Integrated with CoMFA 3D QSAR predictive models, molecular surface property (electrostatic and steric) mapping and structure-based (both ligand and receptor) virtual screening have been employed to explore potential novel hits for the HMGR inhibitors. A representative set of eight new compounds of non-statin-like structures but with high pIC(50) values were sorted out in the present study.

摘要:
A novel solid phase extraction sorbent, nanometer titanium dioxide immobilized on silica gel (immobilized nanometer TiO2), was prepared by sol-gel method, and the adsorptive potential of immobilized nanometer TiO2 for inorganic arsenic species (As(III) and As(V)) was assessed. It was found that both As (III) and As(V) could be adsorbed quantitatively on immobilized TiO2 within a pH range of 5.0-7.5, and only As(III) could be quantitatively retained on the sorbent within a pH range of 9.5-10.5 while As(V) was passed through the microcolumn without the retention. Based on this fact, a new speciation scheme for inorganic arsenic was established, which involved determining total As at pH 6.0 and As(III) at pH 10, with As(V) obtained by difference. The adsorption capacity of immobilized nanometer TiO2 for As(III) was found to be 4.22 mg g-1. Under the optimized conditions, the detection limits of this method for As(III) were 24 ng L-1 with an enrichment factor of 50, and the relative standard deviation (R.S.D.%) was 4.8% (n = 11, c = 5 ng mL-1). The proposed method has been applied to the speciation of inorganic arsenic in natural water samples with satisfactory results. © 2007 Elsevier B.V. All rights reserved.

摘要:
A series of new chromone analogues bearing heterocyclic thioether moiety and aurone analogues bearing cyclic tertiary amine moiety were designed and synthesized under microwave irradiation. The synthetic protocol was found to present many advantages, such as higher yields, shorter reaction time (10-20 min), mild condition, and readily isolation of the products. The synthesized compounds were assayed for their antitumor activity against four kinds of human solid tumor cell lines including HCCLM-7, Hep-2, MDA-MB-435S, and SW-480. Two compounds, (Z)-2-((4-benzyl-piperazin-1-yl)methylene)benzofuran-3(2H)-one 5e and (Z)-2-((4-(bis(4-fluorophenyl)methyl)piperazin-1-yl)methylene)benzofuran-3(2H)-one 5f, were identified as the most promising candidates with the IC50 values in the range of 4.1-13.1 μM. Further cell cycle studies revealed that compounds 5e and 5f arrest the cell cycle in G0/G1 phase and displayed apoptosis-inducing effect on Hep-2 cells. © 2007 Elsevier Ltd. All rights reserved.

摘要:
A species of bacteria that is capable of utilizing imazaquin as the sole carbon source was isolated from soil with repeated imazaquin applications, and was identified as Arthrobacter crystallopoietes (designated as strain "WWX-1"). This isolate degrades imazaquin as high as 200 μg ml<sup>-1</sup>, and the estimated dissipation half-lives increased from 1.51 d for the treatment at 50 μg ml<sup>-1</sup> to 4.75 d for 200 μg ml<sup>-1</sup>. Optimal growth of WWX-1 in mineral salt medium with 50 μg ml<sup>-1</sup> imazaquin was obtained at 35 °C and a pH of 5.0. Growth of WWX-1 was also observed in mineral salt medium with the addition of other imidazolinone herbicides such as imazethapyr and imazapyr, but not with different classes of herbicides such as metsulfuron-methyl. Two imazaquin metabolites were detected, and spectral analysis with HPLC-MS, <sup>1</sup>H NMR, and IR revealed one metabolite with a molecular weight (MW) of 199 as quinoline-2,3-dicarboxylic anhydride. We propose that A. crystallopoietes (WWX-1) could serve as an efficient biodegradation system for remediation of water and soils that are heavily contaminated with imazaquin or other structurally similar chemicals. ©2006 Elsevier Ltd. 用荧光光谱和紫外吸收光谱法研究了五氯酚对牛血清白蛋白(BSA)的荧光猝灭作用.结果表明,五氯酚可以有规律地使BSA内源荧光猝灭,其猝灭机理可认为是五氯酚与BSA形成复合物的静态猝灭;并获得了不同温度下,五氯酚与BSA作用的结合常数和热力学参数,根据所得结果可推断五氯酚与BSA的作用力为疏水作用力和静电作用力,同时由Forster非辐射能量转移理论计算得出了五氯酚与BSA结合位置的距离。

摘要:
A series of 2-substituted-8-(4,6-dimethoxypyrimidin-2-yloxy)-4- methylphthalazin-1-one derivatives, 7a-7w, were designed via an ortho-substituent cyclization strategy to discover a new herbicidal lead structure. These compounds were synthesized by a seven-step route using 3-hydroxy-acetophenone as a starting material. Determination of the K i values against wild-type A. thaliana acetohydroxyacid synthase (AHAS) (EC 4.1.3.18) indicated that some of the compounds displayed good enzyme inhibition activity comparable to that of KIH-6127. The further preliminary bioassay data on weeds showed that the synthesized compounds exhibited typical injury symptoms of AHAS-inhibiting herbicides, and some of them showed broad-spectrum and high herbicidal activities in postemergence treatments against Echinochloa crusgalli, Digitaria sanguinalis, Setaria viridis, Brassica juncea, Amaranthus retroflexus, and Chenopodium album at an application rate of 150 g ai/ha. To our knowledge, this is the first report of methylphthalazin-1- one derivatives as AHAS inhibitors. © 2006 American Chemical Society.

The carbodiimides 2, obtained from aza-Wittig reactions of iminophosphorane 1 with aromatic isocyanates, reacted with hydrazine to give selectively 6-amino-7H-1,2,3-triazolo[4,5-d]pyrimidin-7-ones 5. Compounds 5 were further transformed to iminophosphoranes 6 by reaction with triphenylphosphine, hexachloroethane and triethylamine. A tandem aza-Wittig reaction of iminophosphorane 6 with isocyanate or acyl chloride generated previously unreported 3,5-dihydro-1,2,3-triazolo[4,5-d]-1,2,4-triazolo[1,5-a]pyrimidin-9- ones 10 or 12 in satisfactory yield. X-ray structure analysis of 10g verified the proposed structure and the reaction selectivity. ©The Royal Society of Chemistry 2006.

The growth performance of malolactic fermenting bacteria Oenococcus oeni NCIMB 11648 and Lactobacillus brevis X2 was assessed in continuous culture. O. oeni grew at a dilution rate range of 0.007 to 0.052 h-1 in a mixture of 5:6 (g l-1) of glucose/fructose at an optimal pH of 4.5, and L. brevis X2 grew at 0.010 to 0.089 h-1 in 10 g l-1 glucose at an optimal pH of 5.5 in a simple and safe medium. The cell dry weight, substrate uptake and product formation were monitored, as well as growth kinetics, yield parameters and fermentation balances were also evaluated under pH control conditions. A comparison of growth characteristics of two strains was made, and this showed significantly different performance. O. oeni has lower maximum specific growth rate (μmax=0.073 h -1), lower maximum cell productivity (Qx max=17.6 mg cell l-1 h-1), lower maximum biomass yield (Yx/smax=7.93 g cell mol-1 sugar) and higher maintenance coefficient (ms=0.45 mmol-1 sugar g-1 cell h-1) as compared with L. brevis X2 (μmax= 0.110 h-1; Qxmax=93.2 g-1 cell mol-1 glucose; Yx/smax= 22.3 g cell mol-1 glucose; ms=0.21 mmol-1 glucose g-1 cell h-1). These data suggest a possible more productive strategy for their combined use in maturation of cider and wine. © Springer-Verlag 2005.

Cloud point extraction (CPE) was applied as a preconcentration step prior to graphite furnace atomic absorption spectrometry (GFAAS) determination of manganese(II) and iron(III) in water samples. After complexation with 1-phenyl-3-methyl-4-benzoyl-5-pyrazolone (PMBP), the analytes could be quantitatively extracted to the phase rich in the surfactant p-octylpolyethyleneglycolphenylether (Triton X-100) and be concentrated, then determined by GFAAS. The parameters affecting the extraction efficiency, such as solution pH, concentration of PMBP and Triton X-100, equilibration temperature and time, were investigated in detail. Under the optimum conditions, preconcentration of 10 ml of sample solution permitted the detection of 0.02 ng ml<sup>-1</sup> of Mn(II) and 0.08 ng ml<sup>-1</sup> of Fe(III) with enrichment factors of 31 and 25 for Mn(II) and Fe(III), respectively. The proposed method was applied to determination of trace manganese(II) and iron(III) in water samples with satisfactory results. ©2006 Elsevier Inc. All rights reserved.

Rhodopsin, the G protein-coupled receptor (GPCR) which mediates the sense of vision, was prepared from calf eyes and used as receptor enriched membrane fraction. In this study it was immobilized onto gold electrode by two different techniques: Langmuir-Blodgett (LB) and a strategy based on a self-assembled multilayer. We demonstrated that Langmuir and LB films of rhodopsin are not stable. Thus, in this study a new protein multilayer was prepared on gold electrode by building up layer-by-layer a self-assembled multilayer. It is composed of a mixed self-assembled monolayer formed by MHDA and biotinyl-PE, followed by a biotin-avidin system which allows binding of biotinylated antibody specific to rhodopsin. The immobilization of rhodopsin in membrane fraction, by the specific antibody bound previously on self-assembled multilayer, was monitored with electrochemical impedance spectroscopy (EIS). In addition, the specificity and sensitivity of this self-assembled multilayer system to the presence of rhodopsin were investigated. No effect was observed when the system was in contact with olfactory receptor I7 in membrane fraction used for control measurements. All these results demonstrate that rhodopsin can be immobilized efficiently, specifically, quantitatively and stably on gold electrode through the self-assembled multilayer. © 2005 Elsevier B.V. A series of Ti<inf>1-x</inf>Zr<inf>x</inf>O<inf>2</inf> solid solutions photocatalysts (x = 0.000, 0.045, 0.090, 0.135, and 0.180) was directly obtained by an ultrasonic spray pyrolysis method. Compared with previous methods for solid solutions, our preparation was very fast. The resulting samples were characterized by X-ray diffraction, scanning electron microscopy, transmission electron microscopy, high-resolution transmission electron microscopy, nitrogen adsorption, and UV-vis diffuse reflectance spectroscopy. The characterizations revealed core-shell spherical structures of the resulting solid solutions. We evaluated photocatalytic activities of the solid solutions on degradation of rhodamine B in aqueous solution under simulated solar light. It was found that Ti<inf>0.91</inf>Zr<inf>0.09</inf>O<inf>2</inf> solid solution exhibited the highest photocatalytic activity among all the as-prepared samples. Its activity was much higher than that of P25. The formation mechanism of core-shell spherical structures was proposed. Moreover, we successfully extended this method to prepare microspheres of ceria and ceria-zirconia solid solutions. We think this general method may be easily scaled up for industrial production of microspherical solid solutions photocatalysts and catalysts. ©2006 American Chemical Society.

摘要:
The use of ester-terminated polyamidoamine dendrimers for stabilizing quantum dots in aqueous solutions, is analyzed. Quantum dots have many advantages in terms of optical properties, such as a narrow, tunable, symmetric emission and photochemical stability in comparison to traditional organic dyes. The key to develop quantum dots as a tool in biological systems is to achieve water solubility, biocompatibility, and photostability. The concentration of surfactant had an important effect on the size of the water soluble quantum-dots nanoparticles. The surface tension of the quantum dots nanoparticles decreased with increasing surfactant concentration. The surfactant could form a layer of hydrophilic film, which could prevent the aggregation of nanoparticles, protect the ester group against hydrolysis, and increase the stability of the quantum dots nanoparticles in aqueous media.

摘要:
A novel open-tubular CEC (OT-CEC) column was prepared by immobilized 5,11,17,23-tetra-tert-butyl-25,27-diethoxy-26,28-dihydroxy-calix[4]arene (Calix[41]) on fused-silica capillary column with sol-gel technology. Calix[4] was initially reacted with γ-glycidoxy-propyltrimethoxysilane (KH-560) to form a new sol-gel precursor (calix[4]-KH-560), and then mixed with another precursor, namely tetraethoxysilane (TEOS). After hydrolysis and condensation, a sol-gei matrix was obtained, in which calix[4] was incorporated. Successful calix[4]-modified sol-gel coating was suggested by infrared (IR) spectra and greatly decreased EOF. In comparison with OT columns prepared by the sol-gel method with TEOS and KH-560 only, the calix[4]-modified soi-gel column showed greatly improved separation of isomeric toluidines, nitrophenois, picolines and neuro-transmitters with structural similarity. Migration time and theoretical plate number reproducibility were satisfactory with RSDs less than 1 and 6% each for within column runs and not more than 3 and 7% each for column-to-column measurements, using toluidine and nitrophenol isomers as test solutes on this column. High separation efficiencies (96 000-300 000 plates/m) for basic toluidine isomers were obtained. This column was also successfully employed to combine extraction procedure for the determination of dopamine (DA) and norepinephrine (NE) in Portulaca oferacea L. The recoveries of DA and NE were 93.3 and 94.2%, respectively. © 2006 Wiley-VCH Verlag GmbH & Co. KGaA, Weinheim.

By using time-dependent density functional theory combined with the polarizable continuum model, a satisfactory assignment of the absorption and circular dichroism spectra and energy transfer flow of the β-subunit in C-phycocyanin (C-PC) was achieved when the protonation of β-84 and β-155 phycocyanobilin (PCB) and their interaction with the protein moiety in C-PC have been taken into account. We attribute the main peak for both β-84 and β-155 as arising from the πelectron excitation of the pyrrole rings and the shoulder peak as arising from the charge transfer from the asparate residue to PCBH<sup>+</sup>. The satisfactory agreement between theory and experiment suggests that Fo¨rster resonance theory prevails such that energy transfer occurs from β<inf>s</inf> (β-155) to β<inf>f</inf> (β-84). ©2006 American Chemical Society.

In KOH, the Mg(II)-bromopyrogallol red (BPR) complex produced a very sensitive polarographic wave at -1.30 V. The wave height was linear with the concentration of Mg(II) in the range of 0.05 to 2 microg/mL. The detection limit was 0.01 microg/mL. The electrochemical behavior of Mg(II)-BPR was studied by electrochemical and spectrophotometric methods. Experiments proved that the polarographic wave of Mg(II)-BPR was due to the reduction of BPR in the Mg(II)-BPR complex. The method, which was sensitive, selective, and simple to perform, was used to determine magnesium in foods, and the results were consistent with those obtained by atomic absorption spectroscopy.

Two hexazinone-degrading bacterial strains were isolated from soil by enrichment culture technique, and identified as Pseudomonas sp. and Enterobacter cloacap, respectively. The two purified isolates, designated as WFX-1 and WFX-2, could rapidly degrade hexazinone with half-lives of 3.08 and 2.95 days in mineral salts medium (hereafter referred to as MSM). In contrast, their mixed bacterial culture (herein abbreviated as MBC) was found to degrade hexazinone, at an initial concentration of 50 mg l<sup>-1</sup>, by enhancing 2.3-fold over that when the isolates were used alone. The degradation of hexazinone by MBC in MSM clearly decreased concomitant with the increase of initial concentration, and the level of hexazinone that was toxic enough to totally inhibit degradation was in the range of 150-200 mg l<sup>-1</sup>. The appropriately combined conditions for hexazinone degradation by MBC in MSM were studied, and found to be pH 5.5, 30°C and at agitation of 120 rpm. The addition of MBC to soil had a greater impact on disappearance of hexazinone, which nearly increased fivefold over that of the control set. As a result, findings in the present investigation provide useful information for soil and water decontamination of hexazinone. ©Springer 2006.

PURPOSE: The release of drug from a polymeric matrix is complicated. It often involves drug diffusion, interface movement and various interactions. It also shows a dependence on the length of polymer rod. With Lc as the 'critical length', three different phases can be distinguished from the kinetic process. Prior to reaching Lc, the interface movement plays a key role on determining the release. Otherwise, the drug diffusion can dominate the release process. Near Lc, however, both factors are involved. The rate of interface movement is closely associated with the time and position in the polymer rod. Taking these characters into account, a common model is presented in this article to be tentatively used to interpret the release process. These results provide preliminarily an insight into the understanding of controlled release process.
