Efficient synthesis of furo[2,3-d]pyrimidin-4(3H)-ones

摘要:
The carbodiimides 4, obtained from reactions of iminophosphorane 3 with aromatic isocyanates, reacted with amines or phenols to give 2-substituted furo[2,3-d]pyrimidin-4(3H)-ones 6 in the presence of catalytic amount of sodium alkoxide or solid potassium carbonate in good yields. ©ARKAT USA, Inc.

摘要:
利用串联Aza-Wittig法合成了两类共计16个未见文献报道的噻吩并吡啶并嘧啶类化合物,通过IR,1H NMR,EI-MS,元素分析等方法对所合成的化合物进行了结构表征,化合物II-1还通过了X单晶衍射的进一步确证.并初步测定了所合成化合物的杀虫、杀菌和除草活性.结果表明:所测化合物的杀虫活性较差,但大部分化合物具有一定的除草活性和良好的杀菌活性.

摘要:
应用芳基异氰酸酯与烯基膦亚胺1的氮杂Wittig反应,得到的碳二亚胺2,再与水合肼作用得到氨基咪唑啉酮衍生物4.而后用4与芳基异氰酸酯(或酰氯)、三苯基膦、六氯乙烷和三乙胺"一锅"反应,得到4,5-二氢咪唑并[1,2-b]-1′,2′,4′-***-4-酮衍生物6或7.探讨了所合成新型稠杂环化合物的生物活性,结果表明部分化合物表现出良好的杀菌活性.如7c在50 mg/L浓度时,对棉花枯萎菌、稻瘟菌、黄瓜灰霉菌和油菜菌核菌的抑制率均达100%.

摘要:
噻吩并嘧啶酮衍生物是一类具有良好生物活性的稠杂环化合物,如一些 2-烷氧基及烷基噻吩并嘧啶酮表现出优良的杀菌活性。另外,***类杀菌剂由于具有杀菌谱广、活性高、可内吸传导、价格合理等优点,一直占据了大量的杀菌剂市场。近年来,新型唑类杀菌剂的研究仍然十分活跃,并开发了一些性能更优良的杀菌剂品种。最近,氮杂 Wittig 反应广泛地应用于氮杂环的合成,该反应原料易得、条件温和、反应选择性好,已成为一种合成氮杂环的有效手段。笔者曾报道应用氮杂 Wittig 反应合成噻吩并嘧啶酮杂环的方法,部分化合物表现出抑菌活性,这里进一步报道应用该法合成含***基的噻吩并嘧啶酮杂环。

摘要:
Abstract 2‐(3H)Benzothiazolethiones (2‐mercaptobenzothiazole) are prepared by an improved method, which utilizes microwave‐assisted cyclization of the corresponding ortho‐haloanlines with potassium O‐ethyl dithiocarbonate. By using microwave irradiation, the relative reactivity of 2‐chloroanilines was greatly improved to the same level as that of 2‐fluoroanilines and 2‐bromoanilines.

摘要:
A one‐pot liquid‐phase combinatorial synthesis of 2‐(4‐oxo‐4H‐1‐benzopyran‐3‐yl)‐4‐thiazolidinones bearing diverse substituents at the 3‐position under microwave irradiation was successfully performed using 3‐formyl chromone, primary amine, and mercaptoacetic acid as reactants. Compared to an identical library generated by conventional parallel synthesis, the microwave‐assisted parallel synthesis approach dramatically decreased the reaction time from an average of 9 h to 5 min, and substantially increased the product yields. The coupling of microwave technology with liquid‐phase combinatorial synthesis constitutes a novel and particularly attractive avenue for the rapid generation of structurally diverse libraries. © 2007 Wiley Periodicals, Inc. In the title compound, C18H14N2, the benzimidazole and naphthalene ring systems are planar, making a dihedral angle of 78.71 (1)° with each other. The packing of the mol-ecules in the crystal structure is mainly due to an inter-molecular N - H⋯N hydrogen bond. © International Union of Crystallography 2007.

摘要:
A library of imidazolin-4-ones has been synthesized using poly(ethylene glycol) (PEG) as soluble polymer support. The imidazolin-4-ones 6 or 7 were synthesized effectively by reaction of primary amine with PEG-supported carbodiimides 4, which were obtained from aza-Wittig reaction of PEG-supported iminophosphoranes 3 with isocyanates. © Georg Thieme Verlag Stuttgart.

摘要:
A series of new 2-substituted 3-(4-chlorophenyl)-5,8,9- trimethylthieno[3′,2′:5,6]pyrido[4,3-d]pyrimidin-4(3H)-ones 8 were synthesized via an aza-Wittig reaction. Phosphoranylideneamino derivatives 6a or 6b reacted with 4-chlorophenyl isocyanate to give carbodiimide derivatives 7a or 7b, respectively, which were further treated with amines or phenols to give compounds 8 in the presence of a catalytic amount of EtONa or K 2CO3. The structure of 2-(4-chlorophenoxy)-3-(4- chlorophenyl)-5,8,9-trimethylthieno[3′,2′:5,6]pyrido[4,3-d] pyrimidin-4(3H)-one (8j) was comfirmed by X-ray analysis. © 2007 Verlag Helvetica Chimica Acta AG.

摘要:
Novel 5-(2-chlorophenylmethylidene)-2-thio-4-imidazolinones 3 and 2-Alkythio-5-(2-chlorophenylmethylidene)-4H-imidazolin-4-ones 4 have been synthesized via a tandem aza-Wittig reaction to develop more imidazolinones that may possess biological activities. The target compounds were identified by GC-MS, IR, and 1H NMR spectroscopies, and element analysis, and, in the case of 4k, its structure was established by single crystal X-ray diffraction. They showed not only fungicidal activities against Gibberella zeae, Cercospora beticola, Fusarium oxysporium, Rhizoctonia solani, and Botryosphaeria berengeriana, but also growth inhibition of Barnyard grass and Cole root and stalk.

The title compound, C12H15N3O, crystallizes with four independent molecules in the asymmetric unit. The five-membered heterocyclic rings are essentially planar. 摘要:
Iminophosphorane 1 reacted with an aromatic isocyanate to unexpectedly give a mixture of carbodiimides 2, 3 and 4 through both the normal and the abnormal aza-Wittig reactions. 3-Aminoimidazolone 10 was obtained from the reaction of hydrazine hydrate with carbodiimide 2. Reaction of 10 with triphenyphosphane, hexachloroethane and triethylamine produced iminophosphorane 11 in good yield. A tandem aza-Wittig reaction of iminophosphorane 11 with isocyanate or CS2 generated 3,5-dihydro-6H-imidazo[1,2-b]-1,2,4-triazol-6-ones 13 or 15 in satisfactory yield. Carbodiimides 18, obtained from normal aza-Wittig reactions of vinyl iminophosphorane 17 with aromatic isocyanates, reacted with hydrazine to give 2-arylamino-3-amino-4H-imidazol-4-ones 20. One-pot reactions of 2-arylamino-3-amino-4H-imidazol-4-ones 20 with isocyanates (or acyl chlorides), triphenyphosphane, hexachloroethane and triethylamine produced 3,5-dihydro-6H-imidazo[1,2-b]-1,2,4- triazol-6-ones 22 (or 23) in good yields. The carbodiimides 2, obtained from aza-Wittig reactions of iminophosphorane 1 with aromatic isocyanates, reacted with hydrazine to give selectively 6-amino-7H-1,2,3-triazolo[4,5-d]pyrimidin-7-ones 5. Compounds 5 were further transformed to iminophosphoranes 6 by reaction with triphenylphosphine, hexachloroethane and triethylamine. A tandem aza-Wittig reaction of iminophosphorane 6 with isocyanate or acyl chloride generated previously unreported 3,5-dihydro-1,2,3-triazolo[4,5-d]-1,2,4-triazolo[1,5-a]pyrimidin-9- ones 10 or 12 in satisfactory yield. X-ray structure analysis of 10g verified the proposed structure and the reaction selectivity. ©The Royal Society of Chemistry 2006.

The thienopyridine derivative 2, obtained from reaction of acetoacetic ester with 1 in the presence of tin tetrachloride, was treated with triphenylphosphine in hexachloroethane and Et3N to give iminophosphorane 3. Iminophosphorane 3 reacted with phenyl isocyanate to give carbodiimide 4, which was further treated with phenols or ethenol to produce 2-substituted 5,8,9-trimethyl-3-phenyl-thieno[3′,2′-5,6]pyrido[4,3- d]pyrimidin-4(3H)-ones 5 in presence of catalytic amount of K2CO 3 or EtONa. The structures of compounds 5 were confirmed by 1H NMR, IR, MS, and elemental analysis.

In the title compound, C19H19N3O, the five-membered imidazolone ring is essentially planar. Intermolecular N - H⋯O hydrogen bonds form an infinite chain. The C - N and C - C bond lengths indicate electron delocalization. © 2006 International Union of Crystallography All rights reserved.

摘要:
The isothiocyanate 3, obtained from aza-Wittig reaction of iminophosphorane 2 with CS2, reacts with phenylhydrazine to give thiosemicarbazide 4. Reactions of 4 with alkyl halides in the presence of K2CO3 directly provided 2-alkylthio-4H-imidazol-4- ones 6 in good yields.

<正>咪唑啉类杂环化合物具有多种生物活性,如除草、杀菌、抗肿瘤。尤其是一些2-烷硫基咪唑啉酮表现出显著的杀菌活性,如罗纳普朗克公司开发的新杀菌剂RPA407213,就是2-甲硫基咪唑啉酮衍生物。由于咪唑啉酮杂环具有优良的生物活性,应用新的合成方法及生物活性的研究一直十分活跃。我们曾应用氮杂Wittig反应合成具有杀菌活性的咪唑啉酮衍生物(Ⅰ)和(Ⅱ).
