脂联素基因单核苷酸多态性T45G与湖北汉族人2型糖尿病的遗传关联研究

摘要:
采用病例.家系对照和随机病例.对照两种设计,分析了603例样本脂联素基因(Adiponectin,APMl)单核苷酸多态性(SNP)rs13061862(T45G)与湖北汉族人群2型糖尿病的相关性.在所有样本中,2型糖尿病病人的G等位基因及GG基因型频率显著高于正常人(G:42.0%比21.7%,P<0.001;GG:13.6%比4.5%,P=0.032);在180个病例.家系对照中,2型糖尿病患者的GG基因型频率显著高于对照组(GG:17.8%比5.6%,P=0.011);在423个随机病例.对照中,2型糖尿病患者GG基因型频率也显著高于对照组(GG:12.2%比3.9%,P=0.025);单因素Logistic回归分析显示,GG基因型是2型糖尿病的危险因子(OR=3.58,95%C/=1.70-7.54).这些结果表明,脂联素基因SNPT45G多态性与湖北汉族人群2型糖尿病的发生发展相关,GG基因型是中国湖北汉族人2型糖尿病的遗传危险因素.

摘要:
对344例2型糖尿病患者和307名正常人PPARGC1A基因单核苷酸多态性rs2970847(Thr394Thr)和rs8192678(Gly482Ser)与2型糖尿病的关系进行了单标记和单体型关联分析以及Logistic回归分析.在单标记分析解中,对照组与病例组Thr394Thr的基因型和等位基因频率有显著差异(基因型,P=0.006;等位基因,P<0.001);Logistic回归分析和单标记分析表明,Thr394Thr的AA基因型及Thr394(ACA)-Ser482单体型增加患2型糖尿病的风险.Gly482Ser的基因型和等位基因频率在对照组与病例组间都无显著差异.认为PPARGC1A基因是湖北汉人的一个2型糖尿病易感基因.

摘要:
通过生物信息学手段分析cyp51基因结构,并根据GenBank登记的玉米黑粉茼cyp51 DNA序列,设计cyp51引物和两对分别截短不同跨膜区的突变体引物,构建了多种重组表达质粒及突变体重组表达质粒.选用不同宿主菌包括Escherichia coli BL21(DE3).BL21(DE3)pLysS和Rosetta(DE3)诱导表达并优化条件.SDS-PAGE分析结果表明:只有突变体pET32-YH-35能够在E. coli BL21(DE3)中高效表达(30℃,0.5 mmol/L IPRG诱导).通过与戊唑醇等4种商品化杀菌剂农药和14种XF系列农药先导化合物的紫外结合光谱分析表明:重组蛋白具有生物学活性.其中一种XF系列化合物的结合常数接近商品化杀菌剂,有可能开发为新的杀菌剂,为设计开发新型高效抗真菌新药提供了理论依据.

摘要:
从湖北省洪湖、仙桃等地采集的活性污泥和土壤中分离得到32株好氧反硝化细菌,对其进行反硝化能力测定,其中3株菌的反硝化能力较强,能以NaN0_3为唯一氮源生长,分别命名为HS-N25,HS-MP12和HS-MP13.这3株菌可以分别在18,15和12 h内将特定培养基SC中起始浓度为10 mmol/L的NO_3-完全降解.通过菌株形态观察、生理生化及16S rDNA分子鉴定,菌株HS-N25,HS-MP12及HS-MP13与蜡状芽孢杆菌(Bacillus cereus)亲缘关系最为接近,同源性达99%.初步鉴定这3株菌为蜡状芽孢杆菌.

摘要:
为探讨邻苯二甲酸丁基苄酯(BBP)对小鼠肥大细胞DNA的损伤作用,采用不同浓度BBP(0、4、20、100μmol·L~(-1))对体外培养的小鼠肥大细胞染毒60min,应用单细胞凝胶电泳法和KCl-SDS沉淀法检测细胞DNA断裂和DNA-蛋白质交联.结果表明,低、中、高浓度(4、20、100μmol·L~(-1))的BBP均可引起小鼠肥大细胞DNA损伤,低浓度BBP引起的DNA损伤主要以DNA断裂为主,而中、高浓度BBP引起的DNA损伤主要以DNA-蛋白质交联为主.

摘要:
邻苯二甲酸二异辛酯[di-(2-ethylhexyl)phthalate,DEHP]是增塑剂的一种,其是否具有毒性一直存在争议,也渐渐引起人们的关注。本实验运用单细胞凝胶电泳技术来研究腹腔注射的邻苯二甲酸二异辛酯[di-(2-ethylhexyl)phthalate,DEHP]对小鼠脑组织细胞DNA的损伤。结果表明DEHP可引起小鼠脑组织细胞DNA的损伤,并且随着DEHP浓度(125、375、500mg/kg)的逐渐增高,DNA的损伤也随之加重。在DEHP的浓度达到125mg/kg和375mg/kg时,其可造成DNA的显著断裂(p<0.01)。综上所述,DEHP可造成小鼠脑组织细胞DNA的损伤,并且存在明显的剂量──效应关系。

HMG-CoA还原酶是甲羟戊酸生物合成的限速酶。系统进化分析表明生物界存在两类HMG-CoA还原酶,Ⅰ类主要存在于真核生物和部分古细菌中,Ⅱ类主要存在于原核生物和少数古细菌中。它汀类药物抑制HMG-CoA还原酶活性,是HMG-CoA还原酶的良好竞争性抑制剂。文章阐述了HMG-CoA还原酶的催化机理及系统进化特征与分类,并比较了两类HMG-CoA还原酶催化特性的差异。

摘要:
Bismuth-film electrode (BiFE) was prepared by ex situ depositing bismuth onto glassy carbon electrode for the detection of organophosphate (OP) pesticides, using methyl parathion (MP) as a model analyte. The sufficiently wide negative potential window available made the BiFE a potentially suitable electrode for application in the field of cathodic electrochemical detection of OPs. The BiFE showed similar or even favorable behavior compared to that of mercury and bare electrode. Operational parameters, including the deposition potential and time of bismuth film, pH of the analytic solution and concentration of surfactant, cetyltrimethyl ammonium bromide (CTAB) have been optimized. Under the optimum experimental conditions, the cathodic voltammetric response was proportional to the concentration of MP range from 3.0 to 100 ng mL-1, with a correlation coefficient of 0.9981. The detection limit was 1.2 ng mL-1 (threefold standard deviation of a blank). Due to the easy preparation and regeneration of BiFE together with its good reproducibility and stability, the combination of the promising BiFE and cathodic voltammetric performances opens new opportunity for fast, simple, and sensitive analysis of OP compounds in samples. © 2008 Elsevier Ltd. 摘要:
目的:探索增塑剂DEHP对小鼠重要脏器脂质过氧化作用的影响,了解DEHP的毒作用机制。
方法:采用腹腔注射的染毒实验技术,给小鼠吸入不同浓度的DEHP,用分光光度法测定小鼠不同脏器中脂质过氧化产物丙二醛(MDA)含量。
结果:(1)在低浓度125mg/kg下,小鼠肝,脑,睾丸发生氧化应激效应,MDA含量极显著低于对照组(P<0.01)(2)在中浓度250mg/kg情况下,肝、脑、睾丸中的MDA含量低于对照组,但高于低浓度组。
结论:低浓度的DEHP对MDA的产生起到抑制作用,但是随着DEHP浓度的增加又出现剂量效应关系。

摘要:
UDP-Glycosyltransferases (UGT) are a large family of enzymes, which catalyze the transfer of a sugar from an activated sugar donor to an acceptor molecule. Both in plants and in mammals, they are important in the maintenance of cellular homeostasis. In this study, two genes (designated GhUGT1 and GhUGT2, respectively) encoding putative UGT were isolated from the cotton fiber cDNA library. The deduced proteins contain the signature sequences of plant UGTs in the C-terminal region. The GhUGT1 gene encodes a polypeptide of 457 amino acids, and displays homology at amino acid levels with the known glycosyltransferase genes. Sequence analysis revealed that the GhUGT2 merely encodes a small protein, as there is a nucleotide substitution that results in formation of a stop codon in its open reading frame. Real-time RT-PCR analysis revealed that the expression of GhUGT1 is higher in the fast growth tissues, such as in fibers and roots. GhUGT2 has also higher expression in roots, but with lower expression levels in fibers and other tissues. The results also showed that the expression of GhUGT1 is higher than GhUGT2. Further study showed that GhUGT1 and GhUGT2 expressions are regulated under osmotic stress, suggesting they may be involved in plants responding to osmotic stress.

摘要:
It is well known that the characteristics of cell lines possibly alter when cell lines are at high-passage number because of the environmental selection. We do not know whether non-permissive or low-permissive cell lines could become permissive or more permissive to virus infection after over-high passage. In the present studies, the alteration of the permissiveness of Spodoptera litura cell line Sl-zsu-1 to three baculovirus infection was investigated after over-high passage, and the possible mechanisms are also investigated. Vigorous apoptosis in Sl-zsu-1 cells was induced by both the recombinant Autographa californica multiple nucleopolyhedrovirus AcMNPV-GFP-actin and the celery looper Anagrapha falcifera multiple nucleopolyhedrovirus AfMNPV, suggesting the replication of the two viruses was blocked by apoptosis. However, the cells infected by S. litura multicapsid nucleopolyhedrovirus SpltMNPV did not undergo apoptosis, but the SpltMNPV titre of the supernatant was not detectable, suggesting this cell line was low-permissive for this virus infection and other factor(s) involved in blockage of the virus replication except apoptosis. However, when Sl-zsu-1 cells had been subcultured continuously for more than 4 years (high-passage cell), which was named as Sl-HP cell line afterwards, no significant apoptosis was induced by the three baculovirus in Sl-HP cells, and many replicated virions or nucleocapsids were observed in the cells. But the permissiveness of Sl-HP cells to the three viruses was very different according to the titre of viruses in the cell cultures. Interestingly, the DNA extracted from SpltMNPV could induce vigorous apoptosis of Sl-HP cells. Altogether, Sl-zsu-1 cell line becomes more permissive to baculovirus infection after over-high passage and multiple paths can block the baculovirus infectivity. © 2008 Springer Science+Business Media B.V.

生长抑素(Somatostatins,SST)对胰腺β细胞胰岛素的分泌有重要的调节作用,这一调节作用与细胞内钙离子浓度变化相偶联.以大鼠胰腺β细胞为研究对象,采用显微荧光测钙技术和膜片钳技术,研究了胞外SST对胞内钙离子信号的影响,初步探讨了其作用机制.结果表明:在细胞外液有钙时,胞外SST可减少由去极化产生的胞外钙离子内流;而在细胞外液无钙时,胞外SST通过动员胞内钙库释放而引起胞浆内钙离子浓度显著增高,并触发胰岛素的分泌.

胰岛素的分泌及其分泌的调控是维持机体内葡萄糖平衡的重要机制,胰岛素分泌量的不足会导致非胰岛素依赖的糖尿病的发生。胰岛素包裹在致密核心大囊泡中,胰腺β细胞通过调控致密核心大囊泡的胞吐过程来调节胰岛素的分泌。胞内Ca^2+浓度是影响胰岛素分泌的重要因素。胰腺β细胞主要通过质膜上的ATP敏感的钾通道、钙通道和胞内钙库的活动改变胞内Ca^2+浓度,从而调控β细胞胰岛素的分泌活动。

Effects of two fertilizers, NH(4)Cl and KCl, on the growth of the edible cyanobacterium Ge-Xian-Mi (Nostoc) and four other cyanobacterial strains were compared at pH 8.3+/-0.2 and 25 degrees C. Their growth was decreased by at least 65% at 10 mmol L(-1) NH(4)Cl but no inhibitory effect was observed at the same level of KCl. Meanwhile, the strains exhibited a great variation of sensitivity to NH(4)(+) toxicity in the order: Ge-Xian-Mi>Anabaena azotica FACHB 118>Microcystis aeruginosa FACHB 905>M. aeruginosa FACHB 315>Synechococcus FACHB 805. The 96-h EC(50) value for relative growth rate with regard to NH(4)(+) for Ge-Xian-Mi was 1.105 mmol L(-1), which was much less than the NH(4)(+) concentration in many agricultural soils (2-20 mmol L(-1)). This indicated that the use of ammonium as nitrogen fertilizer was responsible for the reduced resource of Ge-Xian-Mi in the paddy field. After 96 h exposure to 1 mmol L(-1) NH(4)Cl, the photosynthetic rate, F(v)/F(m) value, saturating irradiance for photosynthesis and PSII activity of Ge-Xian-Mi colonies were remarkably decreased. The chlorophyll synthesis of Ge-Xian-Mi was more sensitive to NH(4)(+) toxicity than phycobiliproteins. Thus, the functional absorption cross section of Ge-Xian-Mi PSII was increased markedly at NH(4)Cl levels >or=1 mmol L(-1) and the electron transport on the acceptor side of PSII was significantly accelerated by NH(4)Cl addition >or=3 mmol L(-1). Dark respiration of Ge-Xian-Mi was significantly increased by 246% and 384% at 5 and 10 mmol L(-1) NH(4)Cl, respectively. 摘要:
Screening interesting biocatalysts directly from soil samples is a more convenient and applicable approach than conventional cultivation-dependent ones. In our present work, a soil-derived metagenomic library containing 24,000 transformants was constructed with an efficient strategy for cloning xylanase genes. A gene encoding the enzyme (XynH) able to hydrolyze xylan was obtained. Similarity analysis revealed that this enzyme is a new member in the family 10 of xylanases. The molecular mass of XynH purified from Escherichia coli was estimated to be 39 kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis analysis. It was found to display the maximal activity at lower temperature, under weakly alkaline conditions, different from most of xylanases. The K m and Vmax values of XynH with birchwood xylan as substrate are 7.5 mg/ml and 190 μmol min-1mg-1, respectively. It is greatly interesting to note that the activity of XynH was not reduced significantly by Mn2+, Zn2+, Co2+, Ag+, and Cu2+, even at the concentration of 5 mM, which strongly inhibits most of the other xylanases studied previously. © 2008 Springer-Verlag.

DNA测序是遗传工程的重要技术之一.自从1977年Sanger发明双脱氧DNA测序方法以来,近20年来DNA测序技术发展很快,经过不断改善的自动化操作已经大大加速了测序的速度,使科学家们测出了几百种生物的基因组.但当前普遍使用的测序仪.一次只能读取l,000个左右的碱基,且DNA样品需要长时间的制各和分析,因而岛速测序技术就显得尤为重要.综述nNA测序技术的发展历程及国际最先进技术Solexa高通量测序技术的原理、流程与优势,并对其应用前景进行了展望.

我国是乙型肝炎的高发区,从人群总体乙型肝炎病毒(HBV)感染率分析,大约10%的儿童也被HBV所感染。而在所有的肝炎病毒中,危害最大的是乙型肝炎病毒。因为感染后,个体可以成为无症状病毒携带者。新生儿期及3岁以下的婴幼儿感染后大多数成为表面抗原或病毒携带者,这些携带者中的一部分能发展成急、慢性乙型肝炎、肝硬化或肝癌。因为肝硬化和肝癌发展病程很长,大约需30-40年,故婴幼儿感染了乙型肝炎病毒并成为病毒携带者,发展成肝硬化和肝癌的可能性要大于成年人期乙型肝炎病毒感染。因此,采取有必要采取措施来尽量防治乙肝的恶化。本文就我国乙型肝炎病毒的研究现状及展望做简要介绍,以供参考。
