天然产物吴茱萸碱一锅法合成的综合实验设计

摘要:
结合天然产物全合成的理论知识,设计了从简单易得的底物出发实现吴茱萸碱一锅法合成的综合化学实验。实验过程包括背景文献调研与分析总结、吴茱萸碱一锅法合成、化合物波谱性质表征以及反应机理研究。该综合实验不仅可以培养学生的文献检索、合成操作以及谱图分析等方面的综合能力,而且可以提高学生对有机合成方法学及反应机理研究的理解,提升学生对科学研究逻辑的认知,为将来从事相关科研或实践工作打下基础。

摘要:
针对当前有机化学实验课程存在重讲授轻实验、重验证轻探究、重结果轻过程等弊端,学生积极性和主动性受抑制等问题,我们将翻转课堂模式应用于有机化学实验的教学中进行探索和实践。改变课前和课中学习方式,课前通过信息技术手段完成知识传授过程,课中师生研讨完成知识内化过程,将教学模式从"以教为主"转变为"以学为主",提高了学生的学习积极性、课堂参与度,培养了学生的自主学习、团队协作、表达交流等能力。我们发现翻转课堂模式下,学生在实验操作失败率、实验产物收率、实验报告质量、理论考试成绩等实验课程综合能力上获得提高,有机化学实验课程教学质量得到进一步提升。

摘要:
A robust, practical, and scalable approach for the construction of 3-substituted 5-chloro-1,6-naphthyridin-4-one derivatives 13 via the addition of Grignard reagents to 4-amino-2-chloronicotinonitrile (15) was developed. Starting with various Grignard reagents, a wide range of 3-substituted 5-chloro-1,6-naphthyridin-4-one derivatives 13 were conveniently synthesized in moderate-to-good yields through addition-acidolysis-cyclocondensation. In addition, the robustness and applicability of this synthetic route was proven on a 100 g scale, which would enable convenient sample preparation in the preclinical development of 1,6-naphthyridin-4-one-based MET-targeting antitumor drug candidates.

摘要:
A 2,7-naphthyridine derivative (AND-OH) was used as a fluorophore in the development of a novel fluorescent turn-on probe (AND-DNBS) by introducing a 2,4-dinitrobenzenesulfonate functionality. This fluorescent probe was successfully used for sensing glutathione (GSH) in aqueous media and showed favorable properties such as a rapid response time (45 s) and a significant Stokes shift (227 nm). The characteristics of the developed probe are better than those of another probe, namely ND-DNBS, which was also synthesized by our group and most other previously reported fluorescent probes for GSH. The AND-DNBS probe was able to detect GSH via a red emission in living A549 cells and zebrafishes. These results show that AND-DNBS has potential applications as an excellent probe for detecting GSH in various environments. © 2020 Elsevier Ltd

摘要:
New N-substituted-3-phenyl-1,6-naphthyridinone derivatives are designed and synthesized, based on structural modification of our previously reported compound 3. Extensive enzyme-based SAR studies and PK evaluation led to the discovery of compound 4r, with comparable c-Met potency to that of Cabozantinib and high VEGFR-2 selectivity, while Cabozantinib displayed no VEGFR-2 selectivity. More importantly, at oral doses of 45 mg/kg (Q.D.), compound 4r exhibits significant tumor growth inhibition (93%) in a U-87MG human gliobastoma xenograft model. The promising selectivity against VEGFR-2 and excellent tumor growth inhibition of compound 4r suggest that it could be used as a new lead molecule for further discovery of selective type II c-Met inhibitors.

摘要:
2,7-Naphthyridine derivatives were developed as fluorophores for the first time to design two fluorescence probes, AND-DNP and ND-DNP, which can be applied for detecting thiophenol in aqueous media. Comparing with ND-DNP, AND-DNP showed more favorable properties such as lower background, larger Stokes shift, and higher fluorescence quantum yield for detecting thiophenol. Moreover, the experimental results were verified by theoretical calculations. Hence, AND-DNP was selected as the superior fluorescence probe to detect thiophenol because of its high sensitivity and selectivity. Based on the experimental results, AND-DNP showed a remarkably larger Stokes shift (225nm), faster response speed (30s) and higher fluorescence enhancement (240-fold) than most other fluorescent probes for thiophenol reported in the literature. For an extended application, AND-DNP was applied to detect thiophenol quantitatively in real water samples. Meanwhile, AND-DNP also detected thiophenol via red emission in living A549cells and zebrafish. All these results proved AND-DNP's potential value as an accurate probe for imaging thiophenol in different environments.

摘要:
The 2,7-naphthyridone scaffold has been proposed as a novel lead structure ofMETinhibitors by our group. To broaden the application of this new scaffold, a series of 8-amino-substituted 2-phenyl-2,7-naphthyridin-1(2H)-one derivatives were designed and synthesized. Preliminary biological screening resulted in the discovery of a new lead of c-Kit and VEGFR-2 kinase inhibitors. Compound 9k exhibited excellent c-Kit inhibitory activity, with an IC50 value of 8.5 nM, i.e., it is 38.8-fold more potent than compound 3 (IC50 of 329.6 nM). Moreover, the compounds 10l and 10r exhibited good VEGFR-2 inhibitory activity, with IC50 values of 56.5 and 31.7 nM, respectively, i.e., they are 5.0-8.8-fold more potent than compound 3 (IC50 of 279.9 nM). Molecular docking experiments provided further insight into the binding interactions of the new lead compounds with c-Kit and VEGFR-2 kinase. In this study, an 8-amino-substituted 2-phenyl-2,7-naphthyridin-1(2H)-one scaffold was identified as the new lead structure of c-Kit and VEGFR-2 kinase inhibitors. © 2019 by the authors.

Thermal treatment of (1H-inden-3-yl)dicyclohexylphosphinium tetrafluoroborate (1) and (3-mesityl-1H-inden-3-yl)dicyclohexylphosphinium tetrafluoroborate (3) with tBuONa followed by [(η 6-cymene) RuCl 2)] 2 in methanol gave the adduct {(η 6- cymene)RuCl 2[(1H-inden-3-yl)PCy 2]} (6) and {(η 6-cymene)RuCl 2[(3-mesityl-1H-inden-3-yl)PCy 2]} (7), respectively. Thermal treatment of (2-phenyl-1H-inden-3-yl) dicyclohexylphosphinium tetrafluoroborate (4) with tBuONa followed by [(η 6-cymene)RuCl 2)] 2 or RuCl 3·3H 2O in methanol gave {Ru[κ(P): (η 6-2-phenyl-1H-inden-3-yl)PCy 2]Cl 2} (8). Whereas (2-mesityl-1H-inden-3-yl)dicyclohexylphosphine (5) reacted with [(η 6-cymene)RuCl 2)] 2 (in toluene) or RuCl 3·3H 2O (in ethanol) to afford {Ru[κ(P):(η 6-2-mesityl-1H-inden-3-yl)PCy 2]Cl 2} (9). The molecular structures of complexes 6, 8 and 9 have been determined by single-crystal X-ray diffraction analysis. In addition, complexes 8 and 9 have been found to catalyze the acceptorless dehydrogenation of alcohols in toluene. 9 displayed high activity and different substrates, including cyclic and linear alcohols, were efficiently oxidized to ketones by using 2.0 mol% of catalyst. © 2012 The Royal Society of Chemistry.

Two new glycoluril derivatives, namely diethyl 6-ethyl-1,4-dioxo-1,2,2a,3, 4,6,7,7b-octa-hydro-5H-2,3,4a,6,7a-penta-aza-cyclo-penta-[cd]indene-2a, 7b-dicarboxyl-ate, C<inf>14</inf>H<inf>21</inf>N<inf>5</inf>O<inf>6</inf>, (I), and 6-ethyl-2a,7b-diphenyl-1,2,2a,3,4,6,7,7b-octa-hydro-5H-2,3,4a,6,7a-penta- azacyclo-penta-[cd]indene-1,4-dione, C<inf>20</inf>H<inf>21</inf>N <inf>5</inf>O<inf>2</inf>, (II), both bearing two free syn-urea NH groups and two ureidyl C=O groups, assemble the same one-dimensional chains in the solid state running parallel to the [010] direction via N - H&mellip;O hydrogen bonds. Furthermore, the chains of (I) are linked together into two-dimensional networks via C - H&mellip;O hydrogen bonds. ©2008 International Union of Crystallography.

摘要:
The title compound, C14H18N4O8, is a glycoluril derivative, composed of two five-membered rings in envelope conformations and two six-membered rings in chair conformations. In the crystal structure, inter-molecular C - H⋯O hydrogen bonds link the mol-ecules into a three-dimensional network. One methyl group and the H atoms on the adjacent C atom are disordered over two positions; the site-occupancy factors are ca 0.62 and 0.38. © International Union of Crystallography 2007.

摘要:
A pair of novel fluorescent molecular clips was synthesized from diethoxycarbonal glycoluril cyclic ether and 1,2-dihydroindazol-3-one. Their structures and conformations were confirmed by single crystal X-ray diffraction and chemosensory properties were studied by fluorescent spectroscope. Both of them exhibited selective recognition toward Fe3+ over other metal ions examined in DMF/CH3OH (50:1, v/v). (c) 2007 Elsevier Ltd. All rights reserved.

The title molecule, C10H10Br4, lies on a crystallographic inversion center which is located at the center of the benzene ring. The crystal structure is stabilized by intermolecular Br⋯Br and C—H⋯Br interactions.

<正>在超分子化学研究中,设计并合成一些能键合中性客体分子的主体分子一直是人们关注的热点.Nolte等小组自1987年来报道了一系列衍生于甘脲的夹状主体分子,它们可以通过氢键,π-π堆积等弱相互作用键合一些芳香客体,这些分子夹受体多数都是通过甘脲衍生物与边墙分子反应得到.这里,我们报道了一种新型的分子夹受体3,直接利用酯基甘脲与边墙分子1一步合成得到分子夹2,分子夹2再与苯炔通过Sonogashi
