摘要:
A scaffold provides a framework and initial support for the cells to attach, proliferate and differentiate, and form an extracellular matrix (ECM) in tissue engineering. Here, xyloglucan (XG) was used as a new synthetic ECM for HepG2 cell attachment in alginate capsules. The effects of XG on HepG2 cells on adherent behavior, albumin secretion, ammonia elimination, cell proliferation and gene expression of Connexin 32 and epithelial-cadherin were investigated. Xyloglucan could also promote the HepG2 cell–matrix interactions and the cell clusters formation of HepG2 cells in three dimensional scaffold, thus enhance the liver-specific functions in the three-dimensional space. © 2014, Springer Science+Business Media Dordrecht.

摘要:
为了探究新型增塑剂邻苯二甲酸二异壬酯(DINP)对小鼠过敏性皮肤炎症的影响,采用皮肤染毒的暴露方式,56只SPF级雄性Balb/C小鼠随机平均分为7组处理40d.用0、1.4、14和140mg/kg DINP对小鼠进行皮肤染毒,并用抗氧化剂褪黑素设置2组来探究氧化应激的介导作用,以及1组生理盐水对照组.染毒结束后取耳组织测定耳肿指数和双耳重量差,并制作耳组织病理学切片,最后检测氧化应激指标.实验结果显示, 14和140mg/kg DINP皮肤暴露组的皮肤炎症有明显加剧(P<0.01),氧化应激指标也有明显变化(P <0.01).这表明,一定浓度的DINP,经较长时间的暴露,对小鼠过敏性皮肤炎症具有明显的佐剂作用.并且,随着DINP浓度升高,小鼠机体氧化应激过度活化,造成机体组织或细胞的氧化损伤,并加剧了炎症反应.

摘要:
New generation biomaterials should be designed to bear long-term antibacterial activity, biocompatibility and even osteogenesis facilitation. In this study, the bifunctional regulation of antibiosis and osteogenesis is realized by the highly-ordered and interconnecting Zn-incorporated ZrO2 nanoarrays, which are prepared by a hydrothermal approach with the precursor hydrolyzed in situ and allow long-term controllable Zn release. The content of incorporated Zn can be easily controlled by pH of the HCl solution used for etching. All Zn-incorporated nanoarrays show good antibacterial properties against both Escherichia coli (E. coli, Gram-negative) and Staphylococcus aureus (S. aureus, Gram-positive), as indicated by high antibacterial rates and the apparent inhibition zone. Analysis of the biocompatibility confirms that the hybrid nanoarrays could cause varying degrees of promotion for the adhesion and spreading of MC3T3-E1 cells. Zn incorporated ZrO2 nanotubes balance antibiosis and osteogenesis delicately, as proved by the up-regulated MTT and ALP activities, as well as the increase of bone-related gene expression (the mRNA level of OCN, OPN and BMP-2). The novel bone implant materials with better antibacterial properties can promote the osteogenesis, and have promising applications in biomedical devices and antibacterial control systems. © The Royal Society of Chemistry 2015.

Much of the research effort focused on the impacts of elevated CO<inf>2</inf>on marine algae but very little work was done on freshwater algae, or on freshwater algal viruses. In this paper, we studied the impacts of elevated CO<inf>2</inf>on the infection of a freshwater cyanobacterium (wild Leptolyngbya sp.) by cyanophage PP that have a wide distribution in China. In a 12-month experiment, logarithmic-phase host cells were infected with cyanophage PP at 370 or 740 μatm pCO<inf>2</inf>concentrations;the burst size, lysing cycle and proportion of adsorption were measured. The results showed that the proportion of adsorption, and burst sizes of cyanophage PP increased significantly with elevated CO<inf>2</inf>concentrations, and the proportion of adsorption increased gradually within the 12 months with the gradual increment of cell width. The result indicated that elevated CO<inf>2</inf>concentration may have significantinfluences on the proliferation dynamics of cyanophage-host systems, and some of the influences may increase gradually in a long-term. ©Kai et al.

New generation bone implants with favourable biocompatibility and long-term antibacterial activity have attracted a great deal of attention due to their bifunctional regulation on osteogenesis and antibiosis, demonstrating great potential for their application in biomedical science. In this study, Zn-incorporated TiO<inf>2</inf>nanoarrays are prepared by a hydrothermal approach and the Zn content in the nanorod-structure scaffolds can be controlled by altering the concentration of the precursor solution, thereby tailoring the Zn incorporated scaffolds to meet the requirements in bifunctional clinical applications. Cytocompatibility of the Zn-incorporated TiO<inf>2</inf>nanoarrays was evaluated by MTT, LDH and ALP assays, and all the results are found to be dependent on both time and Zn content. Among TiO<inf>2</inf>-Zn nanoarrays, TiO<inf>2</inf>-Zn0.2 has the most remarkably stimulative effects on MC3T3-E1 cells compared to untreated controls. Meanwhile, the antibacterial tests confirm that the hybrid nanoarrays can exert inhibitory effects on Escherichia coli (E. coli, Gram-negative) and Staphylococcus aureus (S. aureus, Gram-positive) to various degrees based on time and Zn content. The bifunctional implant materials that allow sustainable Zn release with both superior biocompatibility and long-term antibacterial properties will hold significant promise for scalable productions and open the horizon for further applications as biomedical devices. ©The Royal Society of Chemistry 2015.

摘要:
Reversibly switchable fluorescent proteins (RSFPs) can be effectively used for super-resolution optical fluctuation imaging (SOFI) based on the switching and fluctuation of single molecules. Several properties of RSFPs strongly influence the quality of SOFI images. These properties include (i) the averaged fluorescence intensity in the fluctuation state, (ii) the on/off contrast ratio, (iii) the photostability, and (iv) the oligomerization tendency. The first three properties determine the fluctuation range of the imaged pixels and the SOFI signal, which are of essential importance to the spatial resolution, and the last may lead to artificial aggregation of target proteins. The RSFPs that are currently used for SOFI are low in averaged fluorescence intensity in the fluctuation state, photostability, and on/off contrast ratio, thereby limiting the range of application of SOFI in biological super-resolution imaging. In this study, we developed a novel monomeric green RSFP termed Skylan-S, which features very high photostability, contrast ratio, and averaged fluorescence intensity in the fluctuation state. Taking advantage of the excellent optical properties of Skylan-S, a 4-fold improvement in the fluctuation range of the imaged pixels and higher SOFI resolution can be obtained compared with Dronpa. Furthermore, super-resolution imaging of the actin or tubulin structures and clathrin-coated pits (CCPs) in living U2OS cells labeled with Skylan-S was demonstrated using the SOFI technique. Overall, Skylan-S developed with outstanding photochemical properties is promising for long-time SOFI imaging with high spatial-temporal resolution. © 2015 American Chemical Society.

摘要:
采用变性梯度凝胶电泳(DGGE)分析技术,对青海农村户用沼气池内细菌及古菌群落结构进行分析.结果表明,沼气池含有丰富的细菌及古菌类群,且泥样间的细菌和古菌的群落结构存在差异.4个户用沼气池发酵系统中,细菌分属于5个门,优势类群为拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)和变形菌门(Proteobacteria).在属分类水平上,属于24个类群,最优势类群为理研菌科佩特里单胞菌属(Petrimonas)、假单胞菌属(Pseudomona)、泰氏菌属(Tissierella)和梭菌属(Clostridium).同时,沼气池发酵系统中古菌主要包括热丝菌属(Thermofilum)、甲烷短杆菌属(Methanobrevibacter)、甲烷囊菌属(Methanoculleus)、产甲烷菌属(Methanogenium),其中,Methanobrevibacter 和Methanogenium是沼气池内最优势的产甲烷菌.说明沼气池的产甲烷途径主要是氢营养代谢类型,水解、酸化过程主要由来自动物消化系统内的细菌完成.

Background: Actinobacillus pleuropneumoniae, a Gram-negative bacterium, is the causative agent of porcine pleuropneumonia, a highly contagious and often fatal disease. Because current vaccines confer limited protection against A. pleuropneumoniae infection, the development of more effective vaccines is urgently required. The identification of immunogenic and protective antigens, such as an outer-membrane lipoprotein, will advance this purpose. Results: Sixty putative lipoproteins were predicted from the genomic sequence of A. pleuropneumoniae using multiple algorithms. Here, we focused on the characteristics of the putative lipoprotein Lip40 from A. pleuropneumoniae strain SLW01 (serovar 1). Lip40 shares sequence similarity with many bacterial lipoproteins, and the structural prediction of Lip40 suggests that it is similar to A. pleuropneumoniae TbpB. The N-terminus of Lip40 contains an interesting tandemly repeated sequence, Q(E/D/P)QPK. Real-time RT-PCR indicated that the expression of lip40 was significantly upregulated at 42°C, at 16°C, and under anaerobic conditions. Recombinant Lip40 (rLip40) produced in Escherichia coli BL21(DE3) was specifically recognized by porcine convalescent serum directed against A. pleuropneumoniae. Lip40 was confirmed to localize at the bacterial outer membrane, and its expression was significantly stimulated when A. pleuropneumoniae was cultured under various stress conditions. Lip40 also protected 75% of mice from fatal virulent A. pleuropneumoniae infection. Conclusions: The immunogenic outer-membrane protein Lip40 is stress responsive, protects mice against infection, and might be a virulence determinant. Further investigation of Lip40 should expedite vaccine development and provide insight into the pathogenesis of A. pleuropneumoniae. © 2015 Hu et al.

摘要:
Salinispora arenicola CNS-205 was a first-isolated obligate marine actinomycete. A gene (sare0357), annotated as ‘‘amino acid adenylation domain’’located on the genome of Salinispora arenicola CNS-205, was cloned and characterized. The recombinant target protein Sare0357 was expressed in E. coli. Sare0357 specifically recognized and activated tryptophan (Trp) and phenylalanine (Phe). The basic kinetic parameters of Sare0357 for Trp were K<inf>m</inf> = 0.04 mM, V<inf>max</inf> = 2.1 μM/min, k<inf>cat</inf> = 14.2 min<sup>−1</sup>, and for Phe were K<inf>m</inf> = 0.03 mM, V<inf>max</inf> = 1.6 μM/min, k<inf>cat</inf> = 10.4 min<sup>−1</sup>. Our data elucidated Sare0357 biological role and biochemical properties as a Trp and Phe-activating adenylation domain. ©2015, Springer Science+Business Media Dordrecht.

摘要:
In this work, cotton-like Fe2O3-reduced graphene oxide (rGO) composites were synthesized by a one-step microwave-assisted hydrothermal method, using ferrous sulfate (FeSO4) precursor in graphene oxide (GO) solution. The as-obtained products were characterized by X-ray diffraction (XRD), field-emission scanning electron microscopy (FE-SEM), and Fourier-transform infrared (FT-IR) spectra. The results indicated that the cotton-like Fe2O3 was successfully anchored on graphene sheets with a homogeneous distribution. To demonstrate the potential applications, the sensor was fabricated by using the cotton-like Fe2O3-rGO composites for detection of NO2 at room temperature. The testing results demonstrate that the sensor exhibited an excellent selectivity and a significant response to NO2 with a concentration lower to 1 ppm. The present study may serve as a facile and green approach for preparing graphene materials modified with metal oxides to improve their sensing performances.

In 2013, field campaigns were performed in the naturally ventilated homes of ten schoolchildren living in Wuhan's urban area, with purpose to ascertain the primary indoor pollutants that have been associated with respiratory and allergic health in children. According to the 2-week monitoring, mean temperature and relative humidity were 19.5°C and 60% in autumn and 12.7°C and 55% in winter, respectively;mean level of carbon dioxide (CO<inf>2</inf>) was 525ppm in autumn and 748ppm in winter. Particulate matter (PM) led to the most severe indoor air pollution with 94% of gravimetric concentrations of PM<inf>2.5</inf>far beyond 75μg/m<sup>3</sup>. Indoor/outdoor (I/O) ratio of PM<inf>2.5</inf>level varied between 0.72 and 1.04;furthermore, statistical analysis proved that indoor PM level was significantly associated with outdoor level (R<sup>2</sup>≥0.93, p<0.001, n=16). Formaldehyde and acetaldehyde in air samples were always at the levels far below the recommended limits, 100μg/m<sup>3</sup>and 48μg/m<sup>3</sup>, respectively;but total volatile organic compounds (TVOCs) were higher than the national standard of 600μg/m<sup>3</sup>in some homes. Furthermore, high levels of di(2-ethylexyl) phthalate (DEHP) and dibutyl phthalate (DBP) were frequently detected in house dust. The results showed that airborne Aspergillus and Penicillium caused some concerns of fungal pollution in autumn. In conclusion, in the homes of the schoolchildren in Wuhan's urban area, airborne PM, and DEHP and DBP in house dust are primary pollutants;sometimes TVOCs also lead to indoor air pollution;in addition, airborne fungal components indicate to be a contributing factor to indoor pollution of concern in warm environment.<br/> ©2015 Elsevier Ltd.

摘要:
研究了不同p H值的改性絮凝剂对不同p H值铜绿微囊藻的絮凝除藻效果.结果表明:不论是对于室内培养的铜绿微囊藻DS,还是对于野外发生的铜绿微囊藻水华,藻液絮凝后的终点p H值均为影响絮凝效果的关键因素,伴随藻液p H值的逐步升高,需要同步调低絮凝剂的p H值并使终点p H值为7.0~7.3才最有利于形成絮凝体;对于高浮力的野生蓝藻水华,需要提高絮凝剂中黏土的密度才能使絮凝体有效沉降.上述结果说明,在使用壳聚糖改性絮凝剂控制铜绿微囊藻水华时,应根据水体p H值及藻生物量而调整絮凝剂配方,且应优先选择在水体p H值和藻生物量较低时进行控藻.

摘要:
研究邻苯二甲酸二异壬酯对小鼠肝细胞的氧化损伤.以昆明小鼠为受试动物,随机分为5组,包括1个阴性对照组、4个邻苯二甲酸二异壬酯染毒组,染毒组按0.5,5,50,500mg/kg4个剂量水平,灌胃染毒小鼠14d.以肝组织匀浆测定活性氧(ROS)、还原型谷胱甘肽(GSH)、丙二醛(MDA)和8-羟基脱氧鸟苷(8-OHdG)的含量;以肝组织细胞测定DNA-蛋白质交联(DPC)系数.随着邻苯二甲酸二异壬酯染毒剂量的升高,肝组织的ROS、MDA、8-OHdG含量和DPC系数逐渐上升,GSH含量逐渐降低,各指标呈一定的剂量-效应关系.染毒剂量为50mg/kg 时,ROS、GSH、8-OHdG含量和DPC系数差异有统计学意义(P< 0.05, P< 0.01);染毒剂量为500mg/kg时,上述指标差异均有统计学(P< 0.05,P< 0.01).同时对小鼠肝组织形态进行光镜观察,结果表明,随着染毒剂量的加大,小鼠肝细胞的病理损伤越严重.较高剂量(>50mg/kg)的邻苯二甲酸二异壬酯能造成小鼠肝组织的氧化损伤.

摘要:
Cotton (Gossypium hirsutum) as one of the most important economic crop in the world is often suffering from biotic and abiotic stresses during its growth seasons. Mitogen-activated protein kinase (MAPK) cascades participate in signal transduction of extracellular stimuli and regulate multiple biotic and abiotic stress responses in plants. In this study, a MAPK gene (GhMPK4) belonging to the group B of MAPK family was identified in cotton. Quantitative RT-PCR analysis showed that expression of GhMPK4 was induced by high salinity and osmotic stresses. Overexpression of GhMPK4 in Arabidopsis significantly enhanced the transgenic plants’sensitivity to salt and osmotic stresses and exogenous abscisic acid (ABA). Under NaCl, mannitol and ABA treatments, the rates of seed germination and cotyledon expansion/greening of the GhMPK4 overexpression transgenic lines were remarkably declined compared with those of wild type, and roots of the GhMPK4 overexpression transgenic seedlings were shorter than those of wild type. Chlorophyll and proline contents in leaves of the GhMPK4 transgenic lines were obviously lower than those in wild type. Growth status of the GhMPK4 transgenic plants was worse than that of wild type when being subjected to drought and salinity stresses. Furthermore, the expression levels of the stress-related genes were altered in the transgenic plants under NaCl, mannitol and ABA treatments, compared with those in wild type. Taken the data together, it is suggested that GhMPK4 as a negative regulator may participate in response to salt and osmotic stresses and ABA signaling through affecting the expression of the stress-related genes in plants. ©2015, Springer Science+Business Media Dordrecht.

三门峡黄河公铁两用大桥主桥采用(84+9×108+84)m连续钢桁结合梁结构,主梁恒载1 030kN/m,墩高60~72m,桥址区水平地震动峰值加速度为0.18g,场地类别为Ⅲ类。为了达到抗震设防目标,主墩墩顶采用双曲面球型减隔震支座,支座球心距7m,摩擦系数0.03。采用MIDAS Civil软件建立全桥有限元模型进行地震动力分析,并根据分析结果进行桥墩与基础验算。验算结果表明:在设计地震作用下,主体结构均保持在弹性工作范围内;在罕遇地震作用下,墩底可能进入塑性屈服阶段,墩柱按延性构件设计。根据地震动力分析结果和桥墩、基础验算结果,确定双曲面支座抗剪栓水平承载力按1.4倍的多遇地震响应值设计。

Engineering nanoparticles (ENPs) have been widely used in many areas in virtue of its special physical and chemical properties. And ENPs are one of the major innovative scientific and economic growth things, which may present a variety of questions for organisms and environment. This paper analyzed the ecotoxicological effect of the nanosized silicon dioxide (Nano-SiO2) and the normal sized silicon dioxide (Nor-SiO2) within 24 hours as its bulk counterpart to Daphnia magna with different concentration. To assess the aquatic ecosystem safety, EC50 value for immobilization and LC50 value for mortality were measured respectively. The results show that the toxicity of Nano-SiO2 has obviously dose-dependent effect but the Nor-SiO2 has not, evidently. The optical microscope and digital camera also been used to observe and record the biological form of Daphnia Magna during the exposure and in this study we found that D. magna can uptake and adsorb Nano-SiO2 but only apparently show uptaking to its bulk counterpart. The conclusions of results suggest that there is potential harm to aquatic environment in using Nano-SiO2, and it should deserve special concern.

Pyruvate dehydrogenase multienzyme complex E1 (PDHc E1) is a potential target enzyme when looking for inhibitors to combat microbial disease. In this study, we designed and synthesized a series of novel thiamin diphosphate (ThDP) analogs with triazole ring and oxime ether moieties as potential inhibitors of PDHc E1. Their inhibitory activities against PDHc E1 were examined both in vitro and in vivo. Most of the tested compounds exhibited moderate inhibitory activities against PDHc E1 (IC<inf>50</inf> = 6.1-75.5 μM). The potent inhibitors 4g, 4h and 4j, had strong inhibitory activities with IC<inf>50</inf> values of 6.7, 6.9 and 6.1 μM against PDHc E1 in vitro and with inhibition rates of 35%, 50% and 33% at 100 μg mL<sup>-1</sup> against Gibberella zeae in vivo, respectively. The binding mode of 4j to PDHc E1 was analyzed by a molecular docking method. Furthermore, the possible interactions of the important residues of PDHc E1 with compound 4j were examined by site-directed mutagenesis, enzymatic assays and spectral fluorescence studies. The theoretical and experimental results are in good agreement and suggest that compound 4j could be used as a lead compound for further optimization, and may have potential as a new microbicide. The terrestrial macroscopic cyanobacterium Nostoc commune exhibits remarkable resistance to desiccation stress. This species synthesizes abundant acidic water stress protein (WSPA) in cells upon desiccation and secretes it into the extracellular polysaccharide sheath upon rehydration. However, our knowledge about its cellular role in stress resistance is still rather limited. In this paper, we first revealed that WSPA also occurred in two other macroscopic cyanobacteria Nostoc flagelliforme and Nostoc sphaeroides, but it is more abundant in N. commune. The N. commune wspa1 gene was then heterologously expressed in Arabidopsis thaliana. Phenotypic observation found that WSPA1 conferred increased tolerance to osmotic stress in transgenic plants. The physiological indexes such as relative electrolyte leakage, malondialdehyde, proline accumulation and the maximal quantum efficiency of Photosystem II, were also improved in transgenic plants upon osmotic stress, compared to wild types. In addition, GFP fluorescence analysis of eGFP::wspa1 transgenic plant showed that WSPA1 was localized in the cytoplasm. Therefore, the role of WSPA revealed by this study mainly represented its intracellular function. In general, our research suggested that WSPA may act as a stress protein and involve cellular osmotic stress resistance.

Nostoc flagelliforme is an edible blue-green alga with herbal and dietary values. Due to the diminishing supply of natural N. flagelliforme and the large investment on the development of its cultivation technology, it is anticipated that artificially cultured N. flagelliforme will soon sustain the market supply. Once this change occurs, the storage-associated quality problem will become the focus of attention for future trade. In this paper, we used a chlorophyll fluorescence parameter, maximum quantum efficiency of Photosystem II (Fv/Fm), and several biomarkers to evaluate the quality of several N. flagelliforme samples. It was found that longer storage times resulted in darker coloured solutions (released pigments) and decreased amounts of chlorophyll a (Chl a) and water-soluble sugars (WSS). Additionally, a higher Fv/Fm value suggests better physiological recovery and quality. In actual application, determination of Fv/Fm would be the first step for evaluating the quality of N. flagelliforme, and the biochemical indexes would serve as good secondary markers. ©2013 Elsevier Ltd. Pyruvate dehydrogenase complex (PDHc) plays a key role in pyruvate decarboxylation, the transformation of pyruvate to acetyl-CoA. In this study, a new assay for measuring PDHc activity has been developed. 3-(4,5- Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) is adopted as the electron acceptor in this method, and the change in absorbance caused by the reduction of MTT by hydroxyethyl-TPP, which is the catalyzed product of pyruvate dehydrogenase (PDH, E1), within a certain period of time is measured to reflect PDHc activity. For further impartial evaluation of the characteristics of this newly developed assay, a series of comparative studies were also conducted with several commonly used assays of PDHc activity, including the conventional spectrophotometric assay of NADH accumulation, the p-iodonitrotetrazolium violet (INT)-coupled assay, the 2,6-dichlorophenolindophenol (2,6-DCPIP) assay, and the potassium ferricyanide assay. Results have proved that the spectrophotometric assay using MTT is highly sensitive and inexpensive with little interference from the sulfhydryl compound. In conclusion, the newly established assay is applicable for activity measurement not only in purified PDH and PDH in purified PDHc, but also in crude PDH solution prepared under certain conditions; hence, the assay can indirectly reflect the activity of PDHc activity.
